Treatment protocol |
For AOM/DSS group of mice, a single intraperitoneal injection of AOM was given at Day 1, followed by three cycles of DSS administration (cycle 1: 2%, Day 8~14; cycle 2: 1.5%, Day 29~33; cycle 3: 1.5%, Day 50~54) in the drinking water. For control group, AOM was replaced by saline, and DSS was replaced by distilled water.

Growth protocol |
6-week-old male ICR mice were maintained in Central Sourth University Animal Facility according to the Institutional Animal Care Guidelines. All animals were housed in plastic cages (4 or 5 mice/cage) with free access to distilled water and a standard diet under specific-pathogen free (SPF) conditions of humidity (50±10%), lightning (12-h light/dark cycle) and temperature (24±0.5℃).

Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45℃ on GeneChip mouse 430 2.0 Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneChip® Scanner 3000.

Description |
Gene expression data from low grade dysplasia lesion in the colorectum of the experimental group at the end of 4 weeks after intraperitoneal injection of AOM.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 500.
