Title |
MDMs with IFN at T2 |
Sample type |
RNA |
Source name |
IFNα2 pre-treated monocytes-derived macrophages
Organism |
Homo sapiens |
Characteristics |
cell type: monocytes-derived-macrophages ifn pre-treatment: yes hiv infection: none time: 2 hours
Treatment protocol |
Purified primary macrophages were treated with 1000 IU/ml of IFNα2, for 18 hours before infection. Pre-treated macrophages were infected with the Bal strain of HIV-1 at an MOI of 1
Growth protocol |
Monocytes were plated into 25cc flasks in RPMI 1640 supplemented with 10% human serum and incubated at 37°C.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated for each condition and time point using the TRIzol method according to the manufacturer’s protocol
Label |
biotin
Label protocol |
100 ng of total RNA were converted to cDNA, which was amplified and transcribed to produce biotinylated cRNA using the Two-Cycle cDNA synthesis kit (Affymetrix, Santa Clara, CA)
Hybridization protocol |
15ug of fragmented cRNA were hybridized to Affymetrix Human Genome U133 Plus 2.0 arrays (Affymetrix, Santa Clara, CA) for 16 h at 45°C with constant rotation at 60 rpm. The arrays were washed and stained using the Affymetrix Fluidics Station 450 (protocol EukGE-WS2v5_450).
Scan protocol |
Arrays were read using the Affymetrix GeneChip Scanner 3000 7G
Description |
Gene expression data from IFNα2 pre-treated macrophages at 2hours MS_2_IFN
Data processing |
The data were analyzed with GC Robust Multi-array Average (GCRMA) for normalization
