Treatment protocol |
Ten-week-old female C57BL/6 mice received 20 mg of BAMLET in 2 x 200 μl PBS (BAMLET) or 2 x 200 μl of PBS alone by daily gavage for ten days. The mice were sacrificed two or five weeks after the end of treatment. Alternatively, mice received 20 mg of BAMLET in 5 ml PBS or PBS alone, daily in the drinking water until sacrifice after eight weeks. Healthy untreated C57BL/6 mice intestines were used as controls.

C57BL/6 mice were obtained from Jackson Laboratories at approximately eight weeks of age.

Extraction protocol |
At sacrifice, bladders were aseptically harvested for mRNA extraction and analysis. Total RNA was extracted (RNeasy Mini kit, Qiagen) according to the manufacturer's instructions.

100 ng of total RNA was amplified, labeled and fragmented using the GeneChip 3´IVT PLUS Kit ( ThermoFisher Scientific). Biotinylated cRNA was prepared according to the standard Affymetrix's protocol.

Hybridization protocol |
6ug of fragmented and labelled cRNA was hybridized onto Mouse Genome 430 PM array strips (Affymetrix) for 16 hours at 45 °C, washed, stained (Applied Biosystems, ThermoFisher Scientific).

Scanning was done using the GeneAtlas systems (Thermo Fisher).

Data processing |
Transcriptomic data was normalized using Robust Multi Average implemented in the Transcriptome Analysis Console software (v.4.0.1.36, Applied Biosystems, ThermoFisher Scientific).
