Treatment protocol Mock-treated cultures included DMSO vehicle at 0.01% final concentration for 12 hours. Gamma-secretase inhibitor (GSI)-treated cultures included compound E (CmpdE; (S,S)- 2-[2-(3,5-Difluorophenyl)-acetylamino]-N-(1-methyl-2-oxo-5-phenyl-2,3-dihydro-1H-benzo[e][1,4]diazepin-3-yl)-propionamide) (Calbiochem) at 1.0uM final concentration for 12 hours.
Growth protocol Cells were cultured in RPMI 1640 media supplemented with 10% fetal bovine serum, sodium pyruvate, pen/strep, and L-glutamine in a humidified 5% CO2 atmosphere at 37C.

Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Label protocol Ten micrograms of total RNA was used to generate biotin labeled cRNA according to standard Affymetrix protocols.
