Title |
Melanocyte 1 |
Sample type |
RNA |
Source name |
melanocyte
Organism |
Homo sapiens |
Characteristics |
cell type: primary melanocyte
Growth protocol |
Melanoma metastases were collected from 82 patients attending the Newcastle Melanoma Unit. The melanomas were cleaned of surrounding tissue, cut into 2-3 mm fragments and stored in vials in liquid nitrogen. Written consent was given by the patients for collection of their samples. Melanocytes were cultured in Medium 154 (Cascade Biologics) and maintained in exponential growth at 37oC and 5% CO2.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using Trizol reagent (Invitrogen, VIC, Australia) and the RNeasy Kit (Qiagen, VIC, Australia) according to the manufacturers’ instructions.
Label |
Biotin
Label protocol |
Biotinylated cRNA were prepared with the Ambion MessageAmp kit for Illumina arrays
Hybridization protocol |
Standard Illumina hybridization protocol
Scan protocol |
Standard Illumina scanning protocol
Data processing |
All samples were cubic spline normalised and background subtracted using GenomeStudio v2009.2 software (Illumina). Two-hundred and forty three probes, identified through literature and database searches to either be regulated by p53 or known to regulate p53 activity, were used for further analysis.
