Title |
BMDM stimulated with cmB16F1 and dexa+IL4, rep2 |
Sample type |
RNA |
Source name |
BMDM stimulated with cmB16F1 and dexa+IL4
Organism |
Mus musculus |
Characteristics |
strain: C57BL/6 tissue: bone marrow cells after a total of 7 days cultivation in vitro stimulation: cmB16+Dexa+IL4
Treatment protocol |
At day 4 the culture medium was replaced by stimulation medium as indicated and cells were cultured for 3 more days.
Growth protocol |
Bone marrow cells were isolated from the tibias and femurs of C57BL/6 mice. M-CSF at a concentration of 30 ng/ml was added to the culture medium [DMEM, 10% FCS, and penicillin/streptomycin].
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA extraction was carried out with a RNeasy RNA-extraction Kit (Qiagen) according to the manufacturers protocols
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol.
Hybridization protocol |
Hybridization (16h x 45°C) was processed according to the standard Affymetrix protocol.
Scan protocol |
Affymetrix GeneArray Scanner3000
Description |
mouse bone marrow cells after a total of 7 days cultivation in vitro stimulated with cmB16F1 and dexa+IL4, rep2
Data processing |
The data were analyzed with a commercial software called JMP Genomics, version 3.2, from SAS. A custom CDF (Mouse4302_Mm_UG, Brainarray Version 9) was used to annotate the arrays. The Raw fluorescence intensity values were normalized applying quantile normalization.
