Embryos were dechorionated in 5% bleach for ~2-3 min, washed with water. Embryos were then imeidately processed for mRNA extraction.

The total RNA from embryos was extraced using Maxwell simply RNA protocol according to the manufacturers protocol. mRNA-seq: polyA mRNA was purified from total RNA in two consecutive rounds of oligo(dT) selection. A
fter RNA fragmentation and reverse transcription sequencing libraries were prepared following Illumina's instructions (see ChIP-seq)
