Animals were of the following genetic background: 129S7/SvEvBrd-Hprtb-m2, F2, 1 backross to C57BL/6N. Mice were weaned at the age of 4 weeks, and afterwards supplied with standard nutrition until sacrification.

Extraction protocol |
Poles were removed from freshly dissected kidneys, followed by immediate snap-freezing. Total RNA was extracted with Trizol reagent, followed by DNAse treatment and purification via RNEasy columns. RNA purity and concentration was assessed with a Bioanalyzer.

Data processing |
The data were normalised using quantile normalisation with BeadStudio Software. Data were exported to Microsoft Excel for applying student´s t-test on analysis groups of Cldn16 -/- and Cldn16 +/+ animals. Data with a p-value of < 0.05 were considered as significant.
