Status Public on Jan 01, 2021
Title RISP_Ad.lib_5
Sample type SRA

Source name Hypothalamus slice
Organism Mus musculus
Characteristics strain: C57BL/6
genotype: wild type
tissue: Hypothalamus
treatment: RISP_Ad.lib
Treatment protocol Hypothalamus slice were collected from 4-week risperidone diet fed mice at at ad litem or pair feed condition where their body weight were not different from control diet fed mice.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted by the Direct-zol RNA Kits (ZYMO) according to manufacturer’s recommendations and validated by the Agilent 2100 Bioanalyzer.
150 bp paired-end reads were sequenced on DNBseq platform in BGI Americas Corporation.

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model DNBSEQ-G400

Description R7461
Data processing We obtained averaged 2.5 million mapped reads per sample with 90% unique mapped efficiency.
The paired-end clean reads were aligned to the mouse reference genome GRCm38_mm10 (release99) using STAR and each gene reads numbers were count by FeatureCounts.
Differential expression gene (DEG) analysis was performed by DEseq2 package in which gene length and dispersion bias was corrected.
The resulting P values were adjusted using the Benjamini and Hochberg’s approach for controlling the false discovery rate (FDR).
Genes with an adjusted P value < 0.05 and logFold > 0.3 found by DESeq2 were assigned as differentially expressed.
Genome_build: mm10
