cells were harvested (1min, 4000 x g, 4°C) and washed one time with 25ml ice cold H2O. The cells were pelleted and the pellets shock frozen in liquid N2. The cells were homogenized using VK05 glass beads (Precellys), and RNA was extracted using Qiagen's RNeasy Mini Kit.

6 µg of fragmented a-RNA were hybridized for 16hr at 45C in a GeneChip hybridization oven 640.

Arrays were scanned using an Affymetrix GeneChip Scanner 3000 7G.

Summarization using RMA excluding all S. pombe probes was carried out using affy R package. Each temperature was processed separately with the control samples of the corresponding batch.
