Title |
retina_time 0_CD133+_donor3 |
Sample type |
RNA |
Source name |
CD133+
Organism |
Homo sapiens |
Characteristics |
tissue: retina
Treatment protocol |
Retina was dissociated to form a single cell population and CD133+ & CD133- cells were isolated using magnetic beads
Growth protocol |
human retina was disected from post mortem eyes (with research consent)
Extracted molecule |
total RNA |
Extraction protocol |
300ng of RNA were isolated from magnetically sorted cells using RNeasy mini® kit according to the manufacturers instructions
Label |
biotin
Label protocol |
GeneChip Whole Transcript (WT) sense target labeling assay manual, version 4 - starting with 300ng RNA and following the Total RNA labeling protocol to prepare targets for GeneChip gene 1.0 ST Arrays(omitting RiboMinus procedure).
Hybridization protocol |
using the genechip hybridisation, wash and stain kit
Scan protocol |
Array washing and staining as described in chp 6 of above manual using the GeneChip Fluidics station 450 (fluidics script FS450-007). Arrays were scanned on a GeneChip scanner 3000 7G using default scanner settings. Affymetrix GeneChip® Command Console® (AGCC) was used to process all Affymetrix arrays through fluidics and scanning.

Description |
DC_3-time0_(HuGene-1_0-st-v1).CEL
Data processing |
Gene level probe set summarisation (signal value), calculation of QC metrics and CHP file generation were performed using GeneChip® Expression Console software using the Robust Multichip Analysis (RMA) algorithm
