Treatment protocol |
To induce tumor formation, mice were inoculated in the bladder with an adenovirus coding for Cre recombinase. To study the potential activity of CM272+CDDP treatment in BC management, mice were treated daily with CM272(5mg/kg/day) intraperitoneally for 30 days plus a once per week 6 mg/kg dose of CDDP.

Following fragmentation, 5,5 ug of fragmented and labeled ss-DNA were hybridized for 16 hr at 45ÂșC at 60 rpm on Hybridization Oven 640 de Affymetrix. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneArray Scanner 7G de Affymetrix

Data processing |
Expression data were normalized, background-and batch corrected using the Guanine Cytosine Count Normalization (GCCN) and Signal Space Transformation (SST), and prior to summarizing using the Robust Multi-Array Average (RMA) algorithm implemented in the Affymetrix Expression Console software. In order to combine the different microarray data sets, possible batch effects were corrected by using the Bioconductor Combat package in R environment (Johnson et al., 2007)
