Primary myoblasts (PriMBs) were isolated from limb muscle of 10-day old mice using the preplating technique and cultivated to about 70% confluency. Chromatin was prepared and ChIP performed according to Arrigoni, L. et al. (2016) Nucleic Acids Res. 55, e67. Libraries were prepared using the NEBNext Ultra DNA library preparation kit (NEB, E7370S). Adapter-ligated fragments were amplified with 12 PCR cycles and sequenced on the Illumina HiSeq 2500 platform.
