Extraction protocol The smooth muscle cells (SMCs) from the internal anal sphincter (IAS), rectal (RSM), and anocycygeus (ASM) smooth muscle were isolated following the protocol by Rattan and Singh, AJP, 2012. miRNA fractions were isolated from purified SMCs from above different groups of smooth muscel tissues were isolated using miRVana miRNA Isolation kits.
Label biotin
Label protocol The FlashTag™ Biotin HSR Labeling Kit was used to label 500 ng RNA sample.
 
Hybridization protocol Affymetrix gene chips, miRNA-1.0 array (Affymetrix, Santa Clara, CA), were hybridized with FlashTag™ Biotin HSR Labeled RNA sample for 18 h.
Scan protocol Chips were scanned on an Affymetrix Gene Chip Scanner 3000, using Command Console Software.
Description miRNA fractions
Data processing Background correction and normalization were done using RMA with GeneSpring V14.5 software (Agilent, Palo Alto, CA, USA). 2-fold (p-value<0.05) differentially expressed gene list was generated. The differentially expressed gene list was loaded into Ingenuity Pathway Analysis (IPA) 8.0 software (http://www.ingenuity.com) to perform biological network and functional analyses.
