Treatment protocol Mice were treated with 5 daily tamoxifen injections before the start of experiment. DNFB was applied dermally on two consecutive days (day 0 and 1) to promote sensitisation. Inflammation was elicited by a single application of DNFB to the ear pinna on day 5. Ears were harvested after 24 hours and digested in collagenase I. FACS was performed to isolate CD31+CD45-Ter119- endothelial cells. Cells were sorted directly into Qiagen RLT Plus lysis buffer.
Growth protocol NICDiEC-OE mice with endothelial expression of constitutively active Notch1 were generated by combining RosaNICD/NICD with CDH5-iCre transgenics.
Extracted molecule total RNA
Extraction protocol RNA was extracted using the Qiagen RNEasy Plus Micro kit and amplified using the Affymetrix WT Pico Amplification kit, according to manufacturers description.
Label biotin
Label protocol Biotinylated cRNA were prepared from 1ng total RNA and amplified according to manufacturers instructions, using the Affymetric GeneChip WT Pico kit.
 
Hybridization protocol Following fragmentation and terminal labeling, 5,5 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix Gene Chip Mouse Gene 2.1 ST array. GeneChips were washed and stained in the Affymetrix GeneCips fluidics station 450.
Scan protocol GeneChips were scanned using the Affymetrix GeneTitan Multichannel instrument.

Data processing RMA-normalised data were obtained using the Affymetrix Expression Console Software.
