Growth protocol |
M82 and IL8-3 plants were grown in a planting bed with culture soil containing slow-release fertilizer (CDU TamagokaseiS555, Co-op Chemical) and granular dolomite (Magutan, Gausukogyo) in a greenhouse, as described previously (Ikeda et al. 2013). Fruits were sampled at 10, 20, 30 DAF, and ripening stage (breaker stage) from 8 plants. Pericarp tissues were frozen in liquid nitrogen and stored at −80°C¬.

Extraction protocol |
Total RNA was extracted from fruit by the SDS–phenol method (Mohammed et al. 2012) and further purified with an RNeasy Plant Mini Kit (Qiagen). The optical density of the RNA was measured to check the quality, and high-quality RNA was subjected to transcriptome analysis using three independent biological replicates.
