Extraction protocol |
Total RNA was isolated using the SV Total RNA Isolation kit (Promega) protocol on the ground tissue, omitting the DNase treatment in the manufacturer’s protocol. Eluted total RNA was treated with RQ1 Rnase-free DNase (Promega). A 65°C, 15 min incubation step inactivated the enzyme, instead of the kit’s Stop Solution. Multiple identical RNA isolations were pooled and concentrated with the (Qiagen) RNeasy (Qiagen) cleanup protocol.
