Sample GSM149808 Query DataSets for GSM149808
Status Public on Dec 13, 2006
Title Haloperidol treated 4weeks AffyGC RMA rep1
Sample type RNA

Source name Right brain hemisphere ,C57BL male mouse,Haloperidol treated,Orally,4 weeks
Organism Mus musculus
Characteristics Tissue: Forebrain (left hemisphere), Strain: C57BL, Gender: Male, Age: 4weeks, Drug treatment: Typical antipsychotic Haloperidol, Dose: 1.6mg/kg/day, Route of treatment: Oral treatment-Drug added to drinking Water, Period of treatment: 4 weeks, Method of sacrificing: cervical dislocation.
Treatment protocol Mouse was housed individually in a separate cage under a 12-h light–dark cycle and permitted food and water ad libitum. A period of acclimatization for two weeks was allowed prior to any experimental procedure. Haloperidol was added to the animal’s drinking water in a concentration equivalent to the highest human clinical dosage (i.e. 1.6 mg/kg/day) and delivered through light protected bottles. The weight and water consumption was measured weekly and the drug concentration in water was adjusted accordingly .At the end of 4 weeks animal was sacrificed by cervical dislocation and the brain was removed, dissected into left and right hemispheres and cerebellum, flash frozen in liquid nitrogen and stored at -80 C for future use.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 8-10 microg total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol Following fragmentation, 15 microg of cRNA were hybridized for 16 hr at 45C on Affymetrix GeneChip Murine Genome U74 Version 2 Set MG-U74A. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using GeneArray Scanner (Agilent Technologies, Foster City, CA)..
Description Gene expression data from the right forebrain of C57BL mouse received Haloperidol oral treatment four weeks.
Data processing Normalized expression values were calculated from probe level data (CEL file) using the function of Robust Multi-Array expression measure using sequence information GC-RMA (Wu et al 2004, www.bepress.com/jhubiostat/paper4).Computational steps were implemented in the R-language. GCRMA and it's associated packages were provided by the open source bioconductor project.
