Cells were plated in 10cm plate at 70% confluency. Cells were washed with PBS and then given DMEM augmented with 300uM palmitate for 24 hours, at which point total RNA was harvested.

Hybridization protocol |
10 ug of fragmented target cRNA was hybridized at 45 degrees C with rotation for 16 hours (Affymetrix GeneChip Hybridization Oven 640) to probe sets present on an Affymetrix HG_U133A 2 array. The GeneChip arrays were washed and then stained (SAPE, streptavidin-phycoerythrin) on an Affymetrix Fluidics Station 450.

Data processing |
The results were quantified and analyzed using GCOS 1.2 software using default values (Scaling, Target Signal Intensity = 500; Normalization, All Probe Sets; Parameters, all set at default values).
