Dmbo mice were backcrossed with C57BL/6JWT to remove the Rd1 mutation. Wild-type and dumbo mice were maintained in a 12-hours light/12-hours dark cycle with free access to food and water. Animals were killed at P15 by cervical dislocation.

Retinas were isolated under a microscope to remove extra retinal tissue and snap-frozen at -80°C. Total RNA was individually isolated from each whole retina and purified using the RNeasy minikit as described by the manufacturer. RNA quantities were assessed by NanoDrop®ND-1000 spectrophotometer

For each sample, 100ng of total RNA were amplified using the WT sense strand Target Labelling kit (Affimetryx). The resulting sense cDNA was fragmented by UDG and APE 1 and biotin-labelled with TdT using the GeneChip® WT Terminal labelling kit (Affymetrix).

| Hybridization protocol |
Affymetrix Mouse Gene 1.0 ST arrays were hybridized according to the protocol described in Affymetrix GeneChip® Expression Analysis Manual (Fluidics protocol FS450_0007).

| Scan protocol |
The arrays were scanned using the GeneChip® Scanner 3000 7G (Affymetrix) and raw data were extracted from the scanned images and analyzed with the Affymetrix Power Tools software package.

Normalized expression signals were calculated from Affymetrix CEL files using RMA normalization method.
