Treatment protocol The cells from each placenta were in standard conditions of 5% carbon dioxide/air until exposure to <1% oxygen conditions (<1% O2/5%CO2/10%H2/balance N2), 8% oxygen conditions (8% O2/5%CO2/10%H2/balance N2) or standard conditions with or without active vitamin D (1α, 25-dihydroxyvitamin D3) (10 nM or 100 nM), while the other half of the cells were exposed to <1% oxygen, 8%, or standard conditions for 24 h with or without active vitamin D.
Growth protocol Cultures were plated at a density of 300,000 cells/cm2 and maintained in Dulbecco's modified Eagle's medium containing 10% fetal bovine serum, 20 mmol/liter HEPES pH 7.4 , penicillin (100 units/ml), streptomycin (100 μg/ml), and fungizone (0.25 μg/ml) at 37°C in a 5% carbon dioxide/air atmosphere.
