Title scRNA-Seq of peripheral CD8 T cells from patients with metastatic melanoma taking ICI therapy.
Organism Homo sapiens
Experiment type Expression profiling by high throughput sequencing
Summary Patients with metastatic melanoma have been treated with the new class of drug named immune checkpoint inhibitors. Single-cell RNA-sequencing was performed in circulating CD8+ T cells isolated by cell sorting from 20 patients who were followed for 6 months. Among them, 11 responded to therapy, whilst 9 did not. Overall, the data obtained by sc-RNA seq showed the presence of MAIT cells, that had higher levels in those who showed a better response to treatment.

Overall design scRNA-Seq of peripheral CD8 T cells obtained from 20 melanoma patients (11 Responders and 9 Non-Responders) before (T0), after first (T1) and second (T2) cycle of ICI therapy. Almost all samples were sequenced in four replicates.
