Title Single Cell analysis of the two Human Hematopoietic Stem Cell Populations

Summary Single cell transcriptomic profiling (sc RNA-seq) of the two Human Hematopoietic Stem Cell Populations: CD34+CD38-CD45RA-CD90+CD49f+ (hereafter CD90+CD49f+) and CD34+CD38-CD45RA-CD90-CD49f+ (hereafter CD90-CD49f+)
 
Overall design Purpose: to compare the single cells transcriptomic profiles of the two Human Hematopoietic Stem Cell Populations: CD90+CD49f+ and CD90-CD49f+ cells in order to find common marker genes.
Methods: single cells (from human cord-blood CD34+ cells) were flow-sorted directly into 6x96-well plates (wells A6-H7) of the two CD90+CD49f+ and CD90-CD49f+ cells; 3 independent sorts (S1, S2, S3); 3 plates per cell type.
Multiplexed Nextera XT was used to generate libraries. Each single cell was barcoded then all libraries were pooled per plated for sequencing.
Conclusions: this study led to the discovery of a group of cells expressing a common cell surface antigen.
