    Chromatin environments surrounding Xist, Airn, and Kcnq1ot1 suggest common mechanisms of PRC engagement and spreading. Our data indicate that lncRNA potency can be tightly linked to lncRNA abundance and that within lncRNA-targeted domains, PRCs are recruited to CGIs via lncRNA-independent mechanisms.

    Study showed that chromosome interactions between the Airn gene body and the Slc22a3 promoter are enriched on the maternal allele because Airn expression represses these interactions on the paternal allele. These interactions are not required to upregulate Slc22a3 expression on the maternal allele, indicating that they are not essential promoter- enhancer interactions; they may serve to place both loci in close promixity.

    The Airn lncRNA does not require any DNA elements within its locus to silence distant imprinted genes.

    In cardiomyocytes, silencing of Airn via siRNAs augmented cell death, vulnerability to cellular stress, and reduced cell migration. It also caused less binding of Igf2bp2 to other mRNAs and reduced translation of Igf2bp2 protein. Airn's binding partners were Igf2bp2 and Rpa1.

    The results excluded a role for spliced and unspliced Airn lncRNA products and for Airn nuclear size and location in silencing Igf2r. Instead, silencing only required Airn transcriptional overlap of the Igf2r promoter, which interferes with RNA polymerase II recruitment in the absence of repressive chromatin.
