Monocyte-derived dendritic cells from young and aged donors

Comparison of dendritic cells (DCs) from young (20 to 27 years) and aged (77 to 87 years) donors. DCs are antigen-presenting cells important in initiating and regulating innate and adaptive immune responses. Results provide insight into the molecular basis of the decline in DC function with age.

Gene Expression Data in Human Monocyte-derived Dendritic Cell (MoDC) from Young and Aged Donors

(Submitter supplied) Dendritic cells (DCs) are major antigen-presenting cells that play a key role in initiating and regulating innate and adaptive immune responses. DCs are critical mediators of tolerance and immunity. The functional properties of DCs changes with age. The purpose of this study was to define the age-associated molecular changes in DCs by gene array analysis using Affymatrix GeneChips. We identified up and down-regulated gene expression changes in DC from aged donors compared to young donors.

microRNA profilling in dendritic cells of systemic lupus erythematosus

(Submitter supplied) Recent studies have shown that alterations in the function of dendritic cells (DCs) are involved in the pathogenesis of systemic lupus erythematosus (SLE). However, the role of DCs participating in SLE remains unclear. We profiled the microRNAs (miRNAs) of LPS-stimulated DCs in SLE patients and found diffentially expressed miRNAs in DCs of such group patients.

Gene expression analysis of chicken dendritic cells (DCs) in response to H9N2 avian influenza virus infection

(Submitter supplied) We analyze the differentially expressed genes (DEGs) at the transcriptome level in chicken DCs infected with H9N2 influenza virus compared to mock infection by high-throughput RNA-sequencing technology, and found that H9N2 influenza virus infection induced a strong innate immune response in chicken DCs, but impaired the antigen-processing and –presenting capacity of this cell,

Role of interleukin-4 in the licensing of dendritic cells for the induction of Th2 responses [A]

(Submitter supplied) T helper type 2 (Th2) responses are crucial for defense against infections by helminths and are responsible for the development of allergic reactions that can lead to severe clinical disorders, such as asthma or anaphylaxis, and ultimately to death. The induction of Th2 responses requires a specific activation process, triggered by specialized dendritic cells (DCs), by which naive CD4+ Th0 cells acquire the capacity to produce Th2 cytokines. more...

Improved Innate and Adaptive Immunostimulation by Genetically Modified HIV-1 Protein Expressing NYVAC Vectors

(Submitter supplied) Attenuated poxviruses are safe and capable to express foreign antigens. Poxviruses are applied in veterinary vaccination and explored as candidate vaccines for humans. However, poxviruses express multiple genes encoding proteins that interfere with components of the innate and adaptive immune response. This manuscript describes two strategies aimed to improve the immunogenicity of the highly attenuated, host-range restricted poxvirus NYVAC: deletion of the viral gene encoding type-I interferon-binding protein and development of attenuated replication-competent NYVAC. more...

Expression data from human monocyte derived dendritic cells infected with adenovirus expressing HIV-1 Vpr

(Submitter supplied) HIV-1 Vpr protein is a multifunctional protein which perturbs human transcriptome and interacts with a number of cellular proteins. In this study, we have attempted to explore the efffects of Vpr on human transcriptome and have identified several genes which are involved in innate immune responses. We used the microarray analysis to elucidate the differnetail expression pattern of differnet genes in human dendritic cells infected with HIV-1 Vpr. more...

microRNA expression profiling of various C57Bl/6 myeloid cell populations

(Submitter supplied) microRNAs are assumed to fine-tune cellular mRNA expression levels predisposing them for the control of cell development and cell fates. However, despite increasing appreciation of the role of miRNAs in controlling myeloid cell functions and some evidence for their contribution in in vitro monocyte differentiation, the in vivo role of specific miRNAs in the development and homeostasis of myeloid cells remains to be investigated. more...

Expression data from in vitro derived dendritic cells generated in the presence of FLT3-L from wt and miR-142-/- BM cells

(Submitter supplied) 2 types of dendritic cells (DCs) can be generated in vitro in the presence of Flt3-L: CD4+ equivalent CD24- DCs and CD8+ equivalent CD24+ DCs. miR-142-/- mice show a severe defect in the generation of CD4+ equivalent CD24- DCs. To understand the underlying mechanism, RNA expression was analyzed by Affymetrix microarray from the 2 in vitro subtypes of DCs derived from miR-142+/+ and miR-142-/- bone marrow cells. more...

IL-10-conditioned dendritic cells elicit innate inflammatory gene products in response to danger signals

(Submitter supplied) IL-10-Conditioned Dendritic Cells, Decommissioned for Recruitment of Adaptive Immunity, Elicit Innate Inflammatory Gene Products in Response to Danger Signals Dendritic cells (DCs) are the professional APCs of the immune system, enabling T cells to perceive and respond appropriately to potentially dangerous microbes, while also being able to maintain T cell tolerance toward self. In part, such tolerance can be determined by IL-10 released from certain types of regulatory T cells. more...

Transcriptomic profiling of the adaptive and innate immune responses of Atlantic salmon to Renibacterium salmoninarum infection

(Submitter supplied) Renibacterium salmoninarum is a Gram-positive, intracellular bacterial pathogen that causes Bacterial Kidney Disease (BKD) in Atlantic salmon (Salmo salar). To identify Atlantic salmon genes responsive to R. salmoninarum pathogen, fish were intraperitoneally injected with R. salmoninarum (BKD) or sterile KDM-2 medium (control). Head kidney samples were collected at 13 days post-infection. Using 44K microarray analysis, transcriptome analysis was performed to identify differentially expressed probes in response to different levels of R. more...

Lactobacillus acidophilus induces virus immune defense genes in murine dendritic cells by a TLR-2 dependent mechanism

(Submitter supplied) Lactobacilli are probiotics that, among other health promoting effects, have been ascribed immunostimulating and virus preventive properties. Certain lactobacilli species have been shown to possess strong IL-12 inducing properties. As IL-12 production depends on the up-regulation of type I interferons, we hypothesized that the strong IL-12 inducing capacity of L. acidophilus NCFM in murine bone marrow derived DC is caused by an up-regulation of IFN-β, which subsequently stimulates the induction of IL-12 and the dsRNA binding toll like receptor (TLR)-3. more...

Gene expression profiling defines ATP as a key regulator of human dendritic cell functions

(Submitter supplied) Extracellular ATP and PGE2 are two cAMP-elevating agents inducing semi-maturation of human monocyte-derived dendritic cells (MoDCs). We have extensively compared the gene expression profiles induced by ATPγS and PGE2 in human MoDCs using microarray technology. At 6 h of stimulation, ATPγS initiated an impressive expression profile compared to that of PGE2 (1125 genes compared to 133 genes respectively) but after 24 h, the number of genes regulated by ATPγS or PGE2 was more comparable. more...
