Gain-of-function model for conditional activation of IKK2/NF-κB in pancreatic beta-cells

Analysis of islets from 11-week-old Inhibitor of κB kinase 2/pancreatic and duodenal homeobox 1 transgenics. The IKK2/Pdx-1 model allows activation of proinflammatory IKK2/NF-κB signaling, specific to β-cells. Results provide insight into molecular consequences of NF-κB activation in β-cells.

Gene expresssion changes in pancreatic islets of 11 weeks old IKK2-CApdx-1 mice compared to control and Pdx-1+/- mice

(Submitter supplied) Canonical IKK/NF-κB signaling is a master regulator of inflammation and innate immunity and has been implicated in the pathogenesis of T1D. To investigate the impact of NF-κB activation on β-cell homeostasis and diabetes development, we generated a transgenic gain-of-function mouse model allowing conditional NF-κB activation via expression of IKK2-CA (constitutively active IKK2 allele) in β-cells using the tetracycline-regulated gene expression system. more...

Ikk2-mediated inflammatory activation of arterial endothelial cells promotes the development and progression of atherosclerosis

(Submitter supplied) Inflammatory activation of endothelial cells is considered to be the first step in the development of atherosclerosis. Here, we determined the consequences of chronic endothelial NF-κB activation on the development and progression of atherosclerosis as well as plaque regression.

Expression data from SOX9 overexpressing EndoC-ßH1 cells

(Submitter supplied) Human ß cell dedifferentiation as a potent mechanism of diabetes is gaining prominence. Several data suggest an upregulation of the transcription factor SOX9, a progenitor and duct cell marker during ß cell dedifferentiation. However, its targets in such cells need more understanding. Here, we overexpressed SOX9 and a constitutively active mutant (VP16-SOX9∆TAD) in Human pancreatic beta EndoC-ßH1 cells in order to understand its targets.

Expression data from PolyIC treated EndoC-ßH1 cells

(Submitter supplied) Type 1 diabetes (T1D) is a chronic disease characterized by an autoimmune-mediated destruction of insulin-producing pancreatic β cells. Environmental factors such as viruses play an important role in the onset of T1D and interact with predisposing genes. Recent data suggest that viral infection of human islets leads to a decrease in insulin production rather than β cell death, suggesting loss of β cell identity. more...

Ikk2 regulates cytokinesis during vertebrate development (Trancriptome profiling from the wild-type and Ikk2 maternal-zygotic mutant zebrafish)

(Submitter supplied) Purpose: The Ikk2 maternal-zygotic mutants are the only vertebrates animals completely depleted globally of the Ikk2 function which is expected to block an activity of the canonical NFkB signaling pathway. Transcriptome profiling of embryos before the midblastula transition (MBT) and after MBT may provide a clean strategy to identify the NFkB target genes. Methods: Zebrafish lines were maintained under standard laboratory procedures. more...
