Malaria resistance

(Submitter supplied) Examination of molecular basis of malaria resistance. Spleens from malaria resistant AcB55 and AcB61strains compared with A/J mice. Animals were sacrificed and spleens were rapidly frozen in liquid nitrogen, and subsequently stored at 80oC. Tissues were mechanically homogenized with a polytron and total cellular RNA was extracted using a commercially available reagent (TMTRIzol; Invitrogen). The poly-(A)+ fraction of the RNA was isolated by chromatography with oligo d(T) cellulose beads. more...

Bleomycin effect on lungs: dose response and time course

Comparison of lungs from C57BL/6J and C3H/HeJ strains, 3 and 6 weeks after treatment with 80, 100, or 125 units of bleomycin/kg. C57BL/6Js are susceptible to bleomycin-induced pulmonary fibrosis, while C3H/HeJs are resistant. Results identify candidate genes underlying fibrosis susceptibility.

Coxsackievirus B3 (CVB3) infection of AJ, B10.A, CSS3 and B6.chr3AJ (heart tissue day 4)

(Submitter supplied) The pathogenesis of viral myocarditis is a multifactorial process involving host genetics, viral genetics and the environment in which they interact. Here, we used a model of infection with Coxsackievirus B3 to characterize the contribution of host genetics to viral myocarditis. We determined heart CVB3 load in mice from a classical intercross between progenitors A/J (H2a) and B10.A-H2a (B10.A) of different genetic backgrounds but with a common H2 haplotype. more...

Expression profiling of liver tissue from C57BL6 x DBA strain

(Submitter supplied) Expression profiling of liver tissue from 111 F2 female mice resulting from a cross between C57BL/6J and DBA/2J inbred mouse strains. Mice were feed a chow diet for 12 months followed by 4 months on an atherogenic high-fat, high-cholesterol diet. Parental and F2 mice were killed at 16 months of age. The livers were removed immediately and flash frozen for expression profiling. See Schadt, et al., Nature , Vol 422(6929):297-302 for details. more...

Gene expression of liver of vaccination-protected mice in response to early patent infections of Plasmodium chabaudi blood-stage malaria

(Submitter supplied) The role of the liver for survival of blood-stage malaria is only fragmentary understood. In the experimental blood-stage malaria Plasmodium chabaudi, protective vaccination induces self-healing and, thus, survival of otherwise lethal infections. This study investigates the response of the liver, in terms of mRNA and lincRNA expression, to vaccination-induced self-healing infections and lethal P. chabaudi malaria at early patency on day 4 p.i., when parasitized erythrocytes begin to appear in peripheral blood. more...

Differential susceptibility to Candida albicans in mice

(Submitter supplied) C5-deficiency in the A/J and recombinant congenic strain, BcA17, is linked to a dysregulated inflammatory response to Candida albicans infection and high morbidity compared to C5-sufficient C57Bl/6J (B6) mice. The recombinant congenic BcA70 strain thus provides an ideal model to study the effect of C5-deficiency on a B6 genetic background. We have used the A/J and BcA70 strains to carry out a detailed analysis of the pathophysiological effects of the uncontrolled inflammatory activity as a result of Candida infection. more...

Cdcs1 a major colitis susceptibility locus in mice; subcongenic analysis reveals genetic complexity

(Submitter supplied) Background and Aims: In the interleukin-10-deficient (Il10-/-) mouse model of IBD, 10 quantitative trait loci (QTL) have been shown to be associated with colitis susceptibility by linkage analyses on experimental crosses of highly susceptible C3H/HeJBir (C3Bir)-Il10-/- and partially resistant C57BL/6J (B6)-Il10-/- mice. The strongest locus (C3Bir-derived cytokine deficiency-induced colitis susceptibility [Cdcs]1 on Chromosome [Chr] 3) controlled multiple colitogenic subphenotypes and contributed the vast majority to the phenotypic variance in cecum and colon. more...
