Growth factor independence 1 knockout effect on LSK hematopoietic stem bone marrow cells

Analysis of Lin-Sca1+ckit+Flt3- (LSKFlt3-) hematopoietic stem cells (HSC) depleted of growth factor independence 1 (Gfi1). Transcriptional repressor Gfi1 restricts HSC proliferation and is essential to self-renewal. Results provide insight into molecular mechanisms underlying Gfi1 effects in HSCs.

Gfi1 regulates survival and lineage commitment of hematopoietic precursors and prevents myeloproliferative diseases

(Submitter supplied) Hematopoietic stem cells (HSCs) and lymphoid-primed multi-potential progenitors (LMPPs) are able to initiate both lymphoid and myeloid differentiation. We show here that the transcriptional repressor Gfi1 (growth factor independence 1) implements a specific gene expression program in HSCs and LMPPs that is critical for their survival and lymphoid differentiation potential. We present evidence that Gfi1 is required to maintain expression of genes involved in lymphoid development such as Flt-3, IL7R, Ebf1, Rag1, CCR9 and Notch1 and controls myeloid lineage commitment by regulating expression of genes such as Hoxa9 or M-CSFR. more...

The Growth factor independence 1 (GFI1) 36N variant predisposes to Acute Myeloid Leukemia (AML) by inducing epigenetic changes at the Hoxa9 locus

(Submitter supplied) GFI136N is a coding Single Nucleotide Polymorphism (SNP) in the gene GFI1 that increases the risk for Acute myeloid leukemia (AML) by 60%. It is present in 3-5% of Caucasians and has a prevalence of 12% among AML patients. We generated knockin mice expressing either the human GFI136N variant or the more common GFI136S form and observed that GFI136N, in contrast to GFI136S, lacked the ability to bind to the Gfi1 target gene and leukemia associated transcription factor Hoxa9 in myeloid precursors and failed to initiate the histone modifications that regulate HoxA9 expression. more...

Effect of GFI1 KD vs GFI1 KO on expression profiling and AML occurence.

(Submitter supplied) In order to indentify the transcriptional differences between GFI1 KD and GFI1 KO mice that may account for differences in occurence of AML between these mice, we performed exresiion profiling by RNA-Seq on healthy young and old mice mice from both genotypes. Expression data from Rb family (Rb, p130 and p107) deficient Hematopoietic stem Cells

(Submitter supplied) Loss of Rb family in HSCs results in a severe phenotype, such as enhanced proliferation and increase in stem cell number. In addition, HSCs were higly mobilized but failed to transplant. Rb family deficient mice rapidly exhibit a myeloproliferative disease with eosinophilic characteristics.
