Heat stress effect on an epithelial cell line

Analysis of HEp2 epithelial cells subjected to heat stress at temperatures of 37 to 40 degrees C. Results provide insight into the regulatory pathways that control the response to heat stress.

Transcriptional Profiles of Human Epithelial Cells in Response to Heat

(Submitter supplied) We hypothesized that broad-scale expression profiling would provide insight into the regulatory pathways that control gene expression in response to stress, and potentially identify novel heat-responsive genes. HEp2 cells were heated at 37 to 43 °C for 60 min to gauge the heat shock response, using as a proxy inducible HSP-70 quantified by western blot analysis. Based on these results, microarray experiments were conducted at 37, 40, 41, 42 and 43°C (3 replicates/temperature x 5 groups = 15 U95Aver2 GeneChips). more...

Thermoprotection in the Drosophila hsf4 mutant

(Submitter supplied) In Drosophila larvae, acquired synaptic thermotolerance following heat shock has previously been shown to correlate with the induction of heat shock proteins (Hsps) including HSP70. We tested the hypothesis that synaptic thermotolerance would be significantly diminished in a temperature-sensitive strain (hsf4) which has been reported not to be able to produce inducible Hsps in response to heat shock. more...

Salmonella Typhimurium: Heat-shocked cells vs. control cells and acid-shocked cells vs. control cells

(Submitter supplied) We investigated if the transcriptional response of S. Typhimurium to temperature and acid variations was hysteretic and showed memory, i.e., if the transcriptional regulation caused by environmental stimuli remained after the stimuli ceased. The transcriptional activity of non-replicating stationary-phase cells of Salmonella Typhimurium caused by the exposure to 45°C and to pH 5 for 30 min was monitored by microarray hybridizations before the application of the stress and at the end of the treatment period, as well as immediately and 30 minutes after conditions were set back to their initial values, 25°C and pH 7, respectively. more...

Impact of HSF1 and HSF2 on gene expression in normal growth conditions and upon proteotoxic stress.

(Submitter supplied) RNA sequencing (RNA-seq) was used to study the impact of heat shock factors 1 and 2 (HSF1 and HSF2) on gene expression in normal growth conditions, upon Bortezomib-induced proteasome inhibition (25 nM, 6 h or 10 h), and upon heat shock (42∘C, 1 h).

Effect of heat shock at 500 atm and pressure shock to 500 atm on Methanocaldococcus jannaschii

(Submitter supplied) Growth and transcriptional profiles of the barophilic methanarchaeon Methanocaldococcus jannaschii were studied at temperatures up to 98C and pressures up to 500 atm. Application of 500 atm of hyperbaric pressure shifted the optimal growth temperature upwards, and heat shock from 88C to 98C at 500 atm resulted in termination of growth. Pressure shock of M. jannaschii from 7.8 to 500 atm over 15-min, the first pressure upshift reported for a barophile, did not accelerate growth. more...
