Histone deacetylase Sirt1 overexpression effect on the heart

Analysis of transgenic hearts overexpressing Sirt1, a class III histone deacetylase. Sirt1 protects against apoptosis and mediates the survival of cardiac myocytes and neurons under stress in vitro. Results provide insight into the in vivo function of Sirt1 in the heart.

Regulation of Gene Expression by Sirt1 in the heart

(Submitter supplied) We harvested the heart from transgenic mice with cardiac specific overexpression of Sirt1 (Tg-Sirt1) and non-transgenic (NTg) control littermate at 3 months of age and then microarray analyses were conducted. Keywords: Gene expression comparison

Age-related gene expression changes in mouse neocortex

(Submitter supplied) Aging is associated with major nuclear changes affecting genomic integrity and gene expression. Here we compare the gene expression profiles in the neocortex of young (5 months old) and old (30 months old) B6xC3 F1 mice. In a related study, we compare genes that change expression with age to SIRT1 associated genes identified by ChIP on chip (NimbleGen) and find a significant overlap, indicating that SIRT1 target genes are preferentially deregulated with age. more...

SIRT1 and H1AcK26 promoter-association in mouse ES cells changes upon oxidative stress

(Submitter supplied) Association of the (histone) deacetylase SIRT1 with promoters was assessed to determine putative SIRT1-regulated genes. Based on the observation that the SIRT1 yeast ortholog Sir2 redistributes across the yeast genome in repsonse to genotoxic stress and double strand breaks (DSBs), we investigated the impact of oxidative stress on the chromatin binding pattern of SIRT1. Oxidative stress causes a major change in SIRT1 binding that is accompanied by an inverse H1K26 acetylation pattern. more...

microRNA and aging

(Submitter supplied) The critical role of the endothelium in governing vascular, tissues homeostasis and pathological processes is increasingly recognized (Deanfield et al., 2007). Cellular senescence of endothelial cells has been proposed to be involved in endothelial dysfunction and atherogenesis (Minamino T et al., 2007), although the mechanisms underlying the aging induced attenuation of endothelium dependent functions are yet to be clarified. more...
