Body-wide genetic deficiency of poly(ADP-ribose) polymerase 14 sensitizes mice to colitis

(Submitter supplied) Inflammatory bowel disease (IBD) is a debilitating and relapsing chronic disease of the gastrointestinal tract affecting millions of people. Here, we investigated the expression and functions of poly (ADP-ribose) polymerase 14 (Parp14), an important regulator of immune cells functions, using a biobank IBD patient cohort as well as the oral dextran sulfate sodium (DSS) exposure mouse IBD colitis, and, in parallel, the oral Salmonella exposure mouse colitis models. more...

MiR-146a alleviates inflammatory bowel disease in mice through systematical regulation of multiple genetic networks

(Submitter supplied) Inflammatory bowel disease (IBD) is a multiple-genes-involved chronic disease and current available targeted drugs for IBD only deliver moderate efficacy. Whether there is a single gene that systematically regulates IBD is not yet known. Here we showed that the expression of miR-146a in colon was elevated in Dextran Sulfate Sodium Salt (DSS)-induced IBD mice and patients with IBD. DSS induced dramatic body weight loss and much more rectal bleeding, shorter colon length and colitis in miR-146a knock-out mice than wild type (WT) mice. more...

Transcriptional reprogramming and resistance to colonic mucosal injury in PARP1-deficient mice

(Submitter supplied) Poly(ADP-ribose) polymerases (PARPs) synthesize and bind branched polymers of ADP-ribose to acceptor proteins using NAD as a substrate, and participate to the control of gene transcription and DNA repair. PARP1, the most abundant isoform, regulates the expression of proinflammatory mediator - cytokines, chemokines and adhesion molecules, and inhibition of PARP1 enzymatic activity reduced or ameliorated autoimmune diseases in several experimental models, including colitis. more...

Profiling of histone 3 lysine 27 acetylation reveals its role in the chronic DSS-induced colitis mouse model

(Submitter supplied) To investigate whether the H3K27ac is involved in the development of inflammatory bowal disease. We established dextran sulfate sodium (DSS)-induced chronic colitis and performed RNA-sequencing and Chromatin Immunoprecipitation followed by NGS sequencing for H3K27ac in the mice colon tissues. We find that the global H3K27ac distribution in colon tissue had no significant difference, while on the typical-enhancers specific in the DSS group, H3K27ac signals were significantly enriched when compared with the control. more...

Transcriptome study of colon epithelial cells in Reg3a-transgenic mice

(Submitter supplied) The human C-type lectin Reg3a (HIP/PAP) is an antimicrobial peptide that kills Gram-positive bacteria. Reg3a preserves gut microbiota homeostasis, reinforces intestinal barrier function and thereby helps to fight induced colitis in mice. Transcriptomic data revealed an upregulation of numerous genes involved in the robustness of the intestinal barrier, and the biosynthesis pathway of mucin core 1 and 3 O-glycans.

Copper Metabolism Domain-Containing 1 Represses Genes That Promote Inflammation and Protects Mice From Colitis and Colitis-Associated Cancer

(Submitter supplied) Background & Aims Activation of the transcription factor nuclear factor-κB (NF-κB) has been associated with the development of inflammatory bowel disease (IBD). Copper metabolism MURR1 domain containing 1 (COMMD1), a regulator of various transport pathways, has been shown to limit NF-κB activation. We investigated the roles of COMMD1 in the pathogenesis of colitis in mice and IBD in human beings.

Protocadherin 20 maintains intestinal barrier function to protect against Crohn’s disease by targeting ATF6

(Submitter supplied) We identified cadherin superfamily member protocadherin 20 (PCDH20) as a crucial factor in CD using transcriptome sequencing. Thus,we elucidated the function and mechanisms of PCDH20 in gut homeostasis, barrier integrity, and CD development.
