Hypothalamic thyroid hormone deficiency underlies reversible anorexia in a mammalian hibernator

(Submitter supplied) Mammalian hibernators survive prolonged periods of cold and resource scarcity by temporarily modulating normal physiological functions, but the mechanisms underlying these adaptations are poorly understood. The hibernation cycle of thirteen-lined ground squirrels lasts for 5–7 months and comprises weeks of hypometabolic, hypothermic torpor interspersed with 24–48-hour periods of an active-like interbout arousal (IBA) state. more...

Neural control of fluid homeostasis is engaged below 10°C in hibernation

(Submitter supplied) Thirteen-lined ground squirrels (Ictidomys tridecemlineatus) hibernate for several months each winter without access to water, but the mechanisms that maintain fluid homeostasis during hibernation are poorly understood. In torpor, when body temperature (TB) reaches 4°C, squirrels decrease metabolism, slow heart rate, and reduce plasma levels of the antidiuretic hormones arginine vasopressin (AVP) and oxytocin (OXT). more...

RNASeq of cold-sensitive and cold-insensitive neurons from the pre-optic area of hypothalamus of mice

(Submitter supplied) The preoptic area of the hypothalamus (POA) contains intrinsically warm and cold-sensitive neurons, which are thought to be critically involved in mammalian thermoregulation. However, the precise physiological roles and the molecular markers of the cold-sensitive POA neurons have not been determined yet. Here, we tackle this problem by performing calcium-imaging guided separation and collection of cold-sensitive and cold-insensitive dissociated neurons from the mouse POA, followed by RNASeq and differential transcriptomics of these cell populations.

Activation of immediate early response genes initiates a restorative gene expression program during each arousal from torpor in hibernating 13-lined ground squirrels

(Submitter supplied) RNA-seq libraries were prepared from replicate samples (n=5-6) from six physiolocial groups to capture key changes in circannual hibernation.

Liver transcriptome dynamics during hibernation are shaped by a shifting balance between transcription and RNA stability

(Submitter supplied) Hibernators dramatically lower metabolism to save energy while fasting for months. Prolonged fasting challenges metabolic homeostasis, yet small-bodied hibernators emerge each spring ready to resume all aspects of active life, including immediate reproduction. The liver is the body’s metabolic hub, processing and detoxifying macromolecules to provide essential fuels to brain, muscle and other organs throughout the body. more...
