Determination of Sigma 54 reglons in Bacillus thuringiensis HD73 strain

(Submitter supplied) To determine Sigma 54 (SigL) reglons in Bacillus thuringiensis HD73 strain, A sigLmutant, HD(ΔsigL::kan), was constructed with insertion of kanamycin resistance gene cassete. We have employed whole genome microarray expression profiling as a discovery platform to identify the difference of gene expression between mutant and wild-type strains.

Expression analysis of Bacillus anthracis Sterne wild-type and deltaClpX mutant strains

(Submitter supplied) Investigation of whole genome expression level changes in Bacillus anthracis Sterne deltaClpX mutant compared to the wild-type strain after growth in nutrient rich media. The deltaClpX mutant used in this study is described in McGillivray et al. 2009. ClpX Protease Contributes to Antimicrobial Peptide Resistance and Virulence Phenotypes of Bacillus anthracis. Journal of Innate Immunity 1(5): 494-506.

GdpS contributes to Staphylococcus aureus biofilm formation by regulation of eDNA release

(Submitter supplied) In Staphylococcus aureus, the role of the GGDEF domain containing protein GdpS remains poorly understood. Previous studies reported that gdpS mutant strains had decreased biofilm formation due to changes in icaADBC expression that were independent of cyclic-di-GMP levels. We deleted gdpS in three unrelated S. aureus isolates, and analyzed the resultant mutants for alterations in biofilm formation, metabolism and transcription. more...

Effect of virX regulator on the expression of chromosomal genes of Clostridium perfringens

(Submitter supplied) The microarrays were subjected to two-color hybridization using Cy5 and Cy3 dyes incorporated in the cDNAs synthesized from total RNA samples of strains. We compared each gene’s transcription level between the wild-type (WT) strain 13 (labeled by Cy5) and the virX-mutant strain (TS186) (labeled by Cy3) on a single DNA microarray.

Pseudomonas aeruginosa two-component regulator BfmR controls bacteriophage lysis and DNA release during biofilm development through PhdA

(Submitter supplied) Biofilms are surface-adhered bacterial communities encased in an extracellular matrix composed of polysaccharides, proteins, and extracelluar (e)DNA, with eDNA being required for the formation and integrity of biofilms. Here we demonstrate that the spatial and temporal release of eDNA is regulated by BfmR, a regulator essential for Pseudomonas aeruginosa biofilm development. The expression of bfmR coincided with localized cell death and DNA release, with high eDNA concentrations localized to the outer part of microcolonies in the form of a ring and as a cap on small clusters. more...
