Airway and parenchyma RNA sequencing in a mouse model of asthma chronic obstructive pulmonary disease overlap

(Submitter supplied) To profile airway and parenchyma transcriptomes in our mouse model of asthma chronic obstructive pulmonary disease overlap, we employed RNA-Seq (TruSeq Stranded mRNA sample preparation, NovaSeq 6000, 20 million 100 bp single reads) as a discovery tool to identify mRNAs of interest in the development of experimental asthma chronic obstructive pulmonary disease overlap. Experimental asthma: Some mice were sensitized and challenged intranasally with house dust mite (HDM) extract 5 days per week for 3 weeks. more...

Function-selective targeting of the extracellular signal-regulated kinase (ERK1/2) mitigates multiple pathophysiological features of allergen-induced asthma in mice

(Submitter supplied) Purpose: To test the in vivo effectiveness of a novel function-selective inhibitor of ERK1/2 in a murine model of house dust mite (HDM)-induced allergic asthma. Methods: Mice challenged with HDM for 3 weeks were treated with SF-3-030 or vehicle and asthma phenotype was assessed by histology and lung function measurements. Total RNA from lung samples were subjected to RNAseq analysis to determine the molecular signature perturbed by SF-3-030 treatment in mice. more...

Effects of endotoxin abundance in house dust mite on the molecular phenotype of inflammation and tissue response in mouse lung

(Submitter supplied) We report that the abundance of endotoxin in commercially available house dust mite extracts significantly alters the lung transcriptome and secretome in a mouse model of allergic asthma. Studies using the HDM model of asthma in mice should consider reporting the abundance of endotoxin in their commercial HDM stocks in an effort to standardize the protocols and to facilitate consistant and reliable comparison between different studies.

Expression data from lungs of Vldlr-/- mice and wild type (WT) B6129SF2/J mice that had received intranasal house dust mite (HDM) challenges to induce experimental asthma or saline, as a control.

(Submitter supplied) The very low density lipoprotein receptor (VLDLR) is a multi-ligand receptor that mediates pleiotropic biological processes, such as brain development. In this dataset, we include the expression data obtained from lungs from mice that had been challenged with house dust mite to induce experimental asthma or saline, as a control. These data are used to obtain genes that are differentially expressed in response to VLDLR signaling.

Very low density lipoprotein receptor knockout effect in house dust mite model of asthma: lung

Analysis of lung from B6129SF2/J animals depleted for very low density lipoprotein receptor (VLDLR) and subjected to intranasal challenge with house dust mite (HDM). VLDLR is a multiligand apolipoprotein E receptor. Results provide insight into the role of VLDLR in HDM-induced airway inflammation.

Transcriptomic analysis of airway macrophages from a murine experimental allergic airway disease model

(Submitter supplied) Airway macrophages (AMs) are the most abundant leukocytes in the healthy airway lumen and have a highly specialised but plastic phenotype that is governed by the local microenvironment. AMs are thought to maintain immunological homeostasis in the steady state, but have also been implicated in the pathogenesis of allergic airway disease (AAD). To better understand these potentially contrasting AM functions, bulk RNA sequencing was performed on murine AMs obtained during experimental AAD driven by repeated house dust mite inhalation (AM[HDM]s), comparing to control AMs from non-allergic mice. more...

Whole lung transcriptomics of a house dust mite model of mild/moderate asthma

(Submitter supplied) Purpose: Identify whole lung gene expression patterns in a house dust mite model of mild/moderate asthma Methods: Lung gene expression profiles of 10 week old BALB/c female mice were generated by ribosome-depleted, 150 nt, paired-end, stranded RNA-seq with Illumina HiSeq v4. Sequence reads that passed quality filters after trimming were analyzed with Sailfish-cir to identify linear RNAs and circular RNAs. more...
