Gene expression profiles in mouse embryonic fibroblasts (MEFs) derived from BCL11B-KO mice

(Submitter supplied) The differentiation of preadipocytes into adipocytes is controlled by several transcription factors, including peroxisome proliferator-activated receptor γ (PPARγ) and CCAAT/enhancer-binding protein α (C/EBPα), which are known as master regulators of adipogenesis. BCL11B is a zinc finger-type transcription factor that regulates the development of the skin and central nervous and immune systems. Here, we found that BCL11B was expressed in the white adipose tissue (WAT), particularly the subcutaneous WAT and that BCL11B−/− mice had a reduced amount of subcutaneous WAT. more...

DMEM treated WT and PKBa -/- MEFs

(Submitter supplied) Mouse embryonic fibroblasts (MEFs) were generated from 13.5-day-old embryos obtained from heterozygous PKBa mice intercrosses (Yang et al., 2003). Briefly, after dissection of head and visceral organs for genotyping, embryos were minced and trypsinized for 30 min at 37°C. Embryonic fibroblasts were then plated and maintained in Dulbecco’s Modified Eagle Medium (DMEM) with 10% foetal calf serum (FCS) (Life Technologies), 100 units/ml of penicillin and 100 mg/ml of streptomycin at 37°C in an atmosphere of 5% CO2. more...

Protein kinase B alpha knockout effect on adipogenesis: time course

Analysis of protein kinase B alpha (PKBalpha) knockout embryonic fibroblasts (MEFs) following treatment with a standard induction cocktail to induce differentiation into adipocytes. Cells examined up to 48 hours following treatment. Results provide insight into the role of PKBalpha in adipogenesis.

LSD1 knockdown in 3T3-L1 preadipocytes

(Submitter supplied) Obesity is often associated with a low-grade systemic inflammation state that contributes to the development of insulin resistance and atherosclerotic complications. This is usually coupled with increased macrophage infiltration in the adipose tissue and a defect in adipocyte differentiation that results in accumulation of hypertrophic fat cells characterized by a deregulated pattern of adipokine expression. more...

Histone demethylase KDM1A deficiency effect on 3T3-L1 preadipocytes: time course

Analysis of undifferentiated 3T3-L1 preadipocytes up to 48hr after siRNA-mediated depletion of Histone demethylase Kdm1a. Knockdown of kdm1a in 3T3-L1 preadipocytes decreases adipogenesis. Results provide insight into the role of kdm1a in adipogenesis.

Genome-wide identification of Bcl11b gene targets

(Submitter supplied) B-cell leukemia/lymphoma 11B (Bcl11b) is a transcription factor showing predominant expression in the striatum. To date, there are no known gene targets of Bcl11b in the nervous system. Here, we define targets for Bcl11b in striatal cells by performing genome-wide expression profiling. Transcriptome-wide analysis revealed that 694 genes were significantly altered in striatal cells over-expressing Bcl11b, including genes showing striatal-enriched expression similar to Bcl11b. more...

Transcriptome analysis of Bcl11b-deficient murine NK cells

(Submitter supplied) Epigenetic landscapes can provide insight into regulation of gene expression and cellular diversity. Here, we examined the transcriptional and epigenetic profiles of seven human blood NK cell populations, including adaptive NK cells. The BCL11B gene, encoding a transcription factor (TF) essential for T cell development and function, was the most extensively regulated, with expression increasing throughout NK cell differentiation. more...

Tcf-1 expression in murine thymus

(Submitter supplied) The DNA binding factor Tcf-1 is one of the most prominently expressed genes in thymocytes yet its global DNA binding pattern remained unknown. Here we have assessed by ChIP-seq the Tcf-1 binding pattern in murine thymocytes in WT B6 mice and in mice expressing a stabilized form of the Tcf-1 binding partner beta-Catenin (CAT mice).

Cell-type specific action of Bcl11b in early T-lineage and group 2 innate lymphoid cell

(Submitter supplied) The zinc finger transcription factor, Bcl11b, is expressed in T cells and group 2 innate lymphoid cells (ILC2s) among hematopoietic cells. In early T-lineage cells, Bcl11b directly binds and represses endogenous E protein antagonist, Id2, expression to prevent pro-T cell from adopting an innate-like fate. In contrast, ILC2s co-express both Bcl11b and Id2. Here, to address this contradiction, we have directly compared Bcl11b action mechanisms in pro-T cells and ILC2s. more...
