Clonal Immortalized Human Glial Cell Lines Support Varying Levels of JC Virus Infection due to Differences in Cellular Gene Expression

(Submitter supplied) JC virus (JCV) is a ubiquitous human polyomavirus that causes the demyelinating disease Progressive Multifocal Leukoencephalopathy (PML). JCV replicates in limited cell types in culture, predominantly in human glial cells. Thus, productive JCV infection is an indicator of the host cell transcription environment. Following introduction of a replication defective SV40 mutant that expressed large T protein into a heterogeneous culture of human fetal brain cells, multiple phenotypes became immortalized (SVG cells). more...

Human polyomavirus JC virus (JCV) infection of clonal immortalized glial SVG cells

Analysis of fetal glial cell line SVG-derived clones 10B1 and 5F4 following infection with JCV. Clone SVG-5F4 shows low levels of viral growth, whereas clone SVG-10B1 is highly permissive to JCV infection. Results provide insight into cell type differences in JCV host range and pathogenesis.

Comparison of gene expression in SVG-A glial cells and SVGR2 glial cells

(Submitter supplied) SVGR2 cells are glial cells which are derived from SVG-A cells. They were created by subjecting SVG-A cells to multiple rounds of lytic infection by the human polyomavirus JCV. SVGR2 cells are the cells that survived this process and are resistant to JCV infection. This experiment was designed to identify gene expression differences that may be responsible for SVGR2 resistance to JCV. Keywords: Cell type

Polyomavirus JCV resistant glial cell line (HG-U133B)

Analysis of SVGR2 glial cells resistant to the human polyomavirus JCV, the causative agent of progressive multifocal leukoencephalopathy. SVGR2 cells are derived from susceptible glial cells (SVG-A). Results provide insight into the molecular mechanisms underlying susceptibility to JCV infection.

Differentiation of human fetal multipotential neural progenitor cells to astrocytes reveals susceptibility factors for JC Virus

(Submitter supplied) Viral infections of the CNS are of increasing concern, especially among immunocompromised populations. Rodent models are often inappropriate for studies of CNS infection, as many viruses, including JC Virus (JCV) and HIV, cannot replicate in rodent cells. Consequently, human fetal brain-derived multipotential CNS progenitor cells (NPCs) that can be differentiated into neurons, oligodendrocytes, or astrocytes, have served as a model for CNS studies. more...
