Lsr2 and its novel paralogue mediate the adjustment of Mycobacterium smegmatis to unfavorable environmental conditions

(Submitter supplied) In this study, we analyze the role of Lsr2 as a transcription factor in M. smegmatis, a saprophytic bacterium whose natural habitat (soil and water) substantially differs from that of the obligatory mycobacterial pathogens. In summary, the combined RNA-seq and ChIP-seq data revealed that Lsr2 controls gene expression either directly by binding their promoter regions or indirectly through DNA loop formation.

Transcriptional profiling of Mycobacterium tuberculosis Rv3852 (hns) knockout mutant

(Submitter supplied) A handful of nucleoid-associated proteins (NAPs) regulate the vast majority of genes in a bacterial cell. H-NS, the Histone-like Nucleoid-Structuring protein, is one of these NAPs and protects Escherichia coli from foreign gene expression. Though lacking any sequence similarity with E. coli H-NS, Rv3852 was annotated as the H-NS ortholog in Mycobacterium tuberculosis, as it resembles human histone H1. more...

Analyses of the GlnR- and AmtR- dependent nitrogen regulatory network of Mycobacterium smegmatis.

(Submitter supplied) Total transcriptome analyses were carried out to investigate changes in transcript patterns in the Mycobacterium smegmatis wild type strain SMR5 due to nitrogen starvation and to expand the knowledge about the role of the two transcriptional regulators of nitrogen metabolism, namely GlnR and AmtR, in these processes. A first experiment revealed enhanced transcript levels of 284 genes and reduced transcripts of 231 genes in the wild type under nitrogen starvation compared to nitrogen surplus. more...

Mycofactocin is associated with ethanol metabolism in Mycobacteria

(Submitter supplied) Mycobacterium tuberculosis (Mtb) assimilates cholesterol during chronic infection, and its in vitro growth in presence of cholesterol requires Mycofactocin (MFT) biosynthesis genes, although the basis of this requirement is unclear. MFT belongs to the class of ribosomally synthesized and post-translationally modified peptides conserved in many Actinobacteria, and its biological function and structure requires further confirmation. more...

The influence of reduced oxygen availability on gene expression in Laboratory (H37Rv) and clinical strains (S7 and S10) of Mycobacterium tuberculosis

(Submitter supplied) Mycobacterium tuberculosis has the ability to persist within the host in a clinically latent stage. One important condition believed to contribute to latency is reduced access to oxygen but the response of M. tuberculosis to hypoxia is partially characterized. Virtually all dormant models against tuberculosis the vaccine tested in animals used laboratory strains H37Rv or Erdman strains. But major outbreaks of TB occur with the strains that have widely different genotypes and phenotypes compare to H37Rv. more...
