Expression data from mouse intestine: C57Bl/6 MTHFR+/- vs BALB/c MTHFR+/-

(Submitter supplied) Previous studies in our laboratory have shown that low folate diet (control diet with 2mg folate/kg, low folate diet with 0.3mg folate/kg) can induce intestinal tumors in BALB/c mice. In addition, we reported that C57Bl/6J mice did not form tumors under the same conditions. We used microarrays to identify the genetic differences between BALB/c and C57Bl/6J mice that promote tumorigenesis in BALB/c mice. more...

Expression data from mouse intestine: BALB/c MTHFR+/+ on control diet vs BALB/c MTHFR+/- on folate deficient diet

(Submitter supplied) Previous studies in our laboratory have shown that low folate diet (control diet with 2mg folate/kg, low folate diet with 0.3mg folate/kg) can induce intestinal tumors in BALB/c mice. We used microarrays to compare MTHFR+/+ BALB/c mice fed control diet and MTHFR+/- BALB/c mice fed low folate diet.

Folate deficiency effect on methylenetetrahydrofolate reductase MTHFR+/- intestinal neoplasia model: duodenum

Analysis of intestine from MTHFR+/- BALB/c mice fed a low folate diet (0.3mg/kg) for 1 yr. The MTHFR C677T polymorphism is associated with risk for colorectal cancer. Results provide insight into mechanisms underlying the impact of folate and MTHFR deficiency on tumorigenesis.

Adipose tissue analysis of wild-type and beta-carotene 15-15'-oxygenase 1 (Bcmo1) ko mice fed under control diet and beta-carotene diet

(Submitter supplied) Cell culture studies indicate that beta-carotene-(BC)-derived apocarotenoid signaling molecules influence the activities of nuclear receptors that regulate many aspects of adipocyte physiology. Here, we analyzed the roles of the two BC metabolizing enzymes, BCMO1 and BCDO2, for this process in mouse models. Wild-type mice converted BC into retinoids and showed reduced body adiposity (by 28%), leptinemia and adipocyte size. more...

Maternal folic acid mild over consumption in mice leads to sex-specific transcriptional alterations in embryonic placenta

(Submitter supplied) Food fortification and increased use of multivitamins have led to concerns over high folate intake periconceptionally. We reported that diets containing 5-fold higher FA than recommended for mice (5xFASD) during pregnancy, resulted in deficiency of methylenetetrahydrofolate reductase (MTHFR) and altered choline/methyl metabolism, with neurobehavioral abnormalities in newborns. Our goal was to determine whether these changes had their origins in the placenta during embryonic development. more...

Testicular MTHFR deficiency may explain sperm DNA hypomethylation associated with high dose folic acid supplementation

(Submitter supplied) Supplementation with high doses of folic acid, an important mediator of one-carbon transfers for DNA methylation, is used clinically to improve sperm parameters in infertile men. We recently detected an unexpected loss of DNA methylation in the sperm of idiopathic infertile men after 6 months of daily supplementation with 5mg folic acid (>10× the daily recommended intake-DRI), exacerbated in men homozygous for a common variant in the gene encoding an important enzyme in folate metabolism, methylenetetrahydrofolate reductase (MTHFR 677C>T). more...

Low paternal dietary folate alters the sperm epigenome and is associated with negative pregnancy outcomes

(Submitter supplied) We hypothesized that the availability of folate, a soluble B vitamin, would alter the levels of DNA methylation in spermatogenesis with consequences for the sperm epigenome and pregnancy outcomes. We fed male mice with either a folate-deficient or a folate-sufficient diet throughout life. Males fed the folate-deficient diet had offspring with increased birth defects, which included craniofacial and musculoskeletal malformations. more...

Effect of folic acid deprivation on on transcriptome upon OTX treatment

(Submitter supplied) Deciphering the impact of metabolic intervention on response to anticancer therapy represents a path toward improved clinical responses. Here, we identify amino acid-related pathways connected to the folate cycle whose activation predicts sensitivity to MYC-targeting therapies in acute myeloid leukemia (AML). We establish that folate restriction and deficiency of the rate-limiting folate-cycle enzyme, MTHFR ― which exhibits reduced-function polymorphisms in about 10% of Caucasians ― enhance resistance to MYC targeting by BET and CDK7 inhibitors in cell lines, primary patient samples and syngeneic mouse models of AML. more...

p16 epimutation modulates the tumor microenvironment and promotes malignancy of Apc-mutant colon cancer

(Submitter supplied) We previously generated a mouse model of p16 epimutation (named as p16cis/cis) through targeted knock-in of a 140-bp Alu-related DNA sequence (cis-element) that facilitates the spread of DNA methylation at promoter CpG islands (CGIs) in cis (Yu et al. JCI, 2014). We generated primary mouse embryonic fibroblasts (MEFs) from p16cis/cis and control mice. By whole genome bisulfite sequencing (WGBS) for comprehensive DNA methylation profiling, we showed that cis-element-mediated approach specifically induces p16 promoter hypermethylation.

Gene repression through epigenetic modulation by PPARA enhances hepatocellular proliferation

(Submitter supplied) Peroxisome proliferator-activated receptor α (PPARA) is a key mediator of lipid metabolism and inflammation. Activation of PPARA in rodents causes hepatocyte proliferation, but the underlying mechanism is poorly understood. This study focused on genes repressed by PPARA and analyzed the mechanism by which PPARA promotes hepatocyte proliferation in mice. Activation of PPARA by agonist treatment was autoregulated, and induced expression of the epigenetic regulator UHRF1 via activation of the newly described PPARA target gene E2f8, that in turn regulates Uhrf1. more...

Chronic Folate Deficiency in the Mouse Hippocampus

(Submitter supplied) Short-term folate deficiency has been linked to cognitive defects. Given folate’s role in regulating nucleotide synthesis and DNA and histone methylation, these changes are often linked to altered gene expression. We hypothesize that chronic folate deficiency will induce memory deficits that can be linked to tissue-specific changes in gene expression. This study placed mice on folic acid deficient or replete diets, containing either no source of folate or AIN-93G normal folic acid intake, beginning post-weaning and persisting through the end of adult life at 18 months. more...
