TNFα and LTβ stimulated UM-SCC 46 gene expression [TNF]

(Submitter supplied) To investigate the regulatory mechanisms and targets of the activation of NF-kB and inflammatory pathways, we treated HPV(-) head and neck cancer line UM-SCC 46 cells with TNFα and LTβ at different time points, and compared the gene expression by microarray. TNFα uniquely induced 172 genes, LTβ specifically induced 202 genes, while 155 genes were induced by both ligands. Total RNA samples were isolated from UM-SCC 46 cells after TNFα or LTβ treatment at different time points, and the gene expression were compared with untreated cell controls.

LIGHT overexpressing T cells and a high fat diet effect on the liver

Analysis of livers of Lck-LIGHT transgenics fed a high fat diet. Lck-LIGHT animals overexpress LIGHT, a tumor necrosis factor cytokine family member, in T cells and develop hyperlipidemia. Results provide insight into the role of T cells and liver in regulating lipid homeostasis mediated by LIGHT.

RNA Helicase DDX5, a Negative Regulator of Wnt Activation and Hepatocyte Reprogramming in Hepatitis B Virus-associated Hepatocellular Carcinoma

(Submitter supplied) We demonstrate that HBV infection induces expression of the proto-oncogenic miR17~92 and miR106b~25 clusters which target the downregulation of DDX5. Increased expression of these miRNAs is also detected in HBV-driven HCCs exhibiting reduced DDX5 mRNA. Stable DDX5 knockdown (DDX5KD) in HBV replicating hepatocytes increased viral replication, and resulted in hepatosphere formation, drug resistance, Wnt activation, and pluripotency gene expression. more...

Gene expression analysis of total B, CD4+ T and epithelial cells in lacrimal glands from the NOD mouse model of Sjogren's syndrome

(Submitter supplied) Despite being one of the most common rheumatologic diseases, there is still no disease-modifying drug for primary Sjögren’s syndrome (pSS). Advancing our knowledge of the target tissue has been limited by the low dimensionality of histology techniques and the small size of human salivary gland biopsies. The goal of this study was to characterize the gene expression profile of total B cells, CD4+ T cells and epithelial cells in the target tissue of the most commonly-used model of pSS (the NOD model of pSS characterized by severe dacryoadenitis developing in males between 8 and 16 weeks of age).

Molecular Signature of Hepatitis D Virus-Associated Hepatocellular Carcinoma

(Submitter supplied) To investigate the role of viral and host factors in HDV-associated HCC we carried out an integrated clinicopathological and gene expression study of tissue specimens and laser microdissected hepatocytes obtained at the time of liver transplantation from livers with HDV-HCC, HDV-cirrhosis without HCC, HCV-HCC and HBV-HCC. References to GSM series of HDV and HBV livers, already deposited in GEO, are included in this series. more...

TNFR1 controls apoptosis and chronic liver disease in hepatocyte-specific IKKγ (Nemo) mice.

(Submitter supplied) Death receptor-mediated hepatocyte apoptosis is implicated in a wide range of liver diseases including viral hepatitis, alcoholic hepatitis, ischemia/reperfusion injury, fulminant hepatic failure, cholestatic liver injury and cancer. Deletion of NF-ĸB essential modulator in hepatocytes (NemoΔhepa) causes the spontaneous development of hepatocellular carcinoma preceded by steatohepatitis in mice and thus serves as an excellent model for the progression from chronic hepatitis to liver cancer. more...

Death receptor knockout effect on NEMO-deficient model of chronic liver disease

Analysis of livers from hepatocyte-specific NF-ĸB essential modulator (NEMO), NEMO-TRIAL, and NEMO-TNFR null males. NEMO deletion in hepatocytes causes spontaneous hepatocyte apoptosis. TNFR1 and TRAIL encode death receptors. Results provide insight into the role of death receptors in liver injury.
