Effects of Arid1a knockout in murine ovarian endometrioid carcinomas with biallelic inactivation of Apc and Pten

(Submitter supplied) We mated mice with floxed alleles of both Apc and Pten, to mice with floxed alleles for Arid1a, to obtain female mice with both copies of all three genes floxed. At 7 to 8 weeks of age the right ovarian bursal cavites of the mice were injected with 50 million plaque-forming units of adenovirus expressing Cre recombinase, which causes the floxed genes to be knocked out. Tumor tissue from 3 mice for each group was obtained at necropsy, RNA purified, and targets for Affymetrix arrays synthesized from the mRNAs. more...

Targets of Wnt-1 in C57MG

(Submitter supplied) RNA from murine mammary C57MG cells transfected with an empty control vector (PLNCx) or an expression vector encoding Wnt-1 was isolated and analyzed on Affymetrix MG-U74Av2 arrays. Sample preparation was done as recommended by the manufacturer. Images were analyzed using Affymetrix Microarray Suite 4.0 software. Comparison of Wnt-1 expressing and control cells revealed Wnt-1 targets. Keywords: ordered

Oviductal vs. ovarian epithelial transformation yields very different tumor phenotypes in Apc fl/fl;Pten fl/fl mice

(Submitter supplied) We treated 6-8 week old mice that had floxed alleles of both Apc and Pten (for both alleles in each case) that also carry an Ovgp1-iCre-ERT2 transgene, with one of two treatments; a third group received neither treatment. The Ovgp1-iCre-ERT2 expresses Cre recombinase fused to a tamoxifen-inducible fragment of the estrogen receptor, in tissues where the Ovgp1 gene (oviductal glycoprotein 1) is expressed, which is almost exclusively in mouse oviductal epithelium (equivalent to human fallopian tube epithelium = FTE). more...

caArray_cho-00156: Gene Expression in Ovarian Cancer Reflects Both Morphology and Biological Behavior

(Submitter supplied) Biologically and clinically meaningful tumor classification schemes have long been sought. Some malignant epithelial neoplasms, such as those in the thyroid and endometrium, exhibit more than one pattern of differentiation, each associated with distinctive clinical features and treatments. In other tissues, all carcinomas, regardless of morphological type, are treated as though they represent a single disease. more...

Gene Expression Profiles of a Progression Model of Human Ovarian Cancer

(Submitter supplied) We have earlier reported the establishment of a comprehensive in vitro panel of 19 isogenic cell lines from a patient with Grade IV serous adenocarcinoma (Bapat et al. Cancer Research. 2005). In this study, one of the cell lines termed as A4 at early passes (less than ~20) was assessed as being non-tumorigenic since it failed to exhibit anchorage independence growth in soft agar and form tumors in nude mice. more...

FGF Receptor and Wnt-1 Signaling Cooperate in Mammary Tumorigenesis

(Submitter supplied) The fibroblast growth factor pathway is known to cooperate with the highly oncogenic Wnt/beta-catenin pathway in mouse models of breast cancer. To investigate the mechanisms involved in this cooperativity, we utilized MMTV-driven transgenic mouse lines expressing a drug-inducible model for FGF Receptor signaling (iFGFR) crossed with the previously characterized MMTV-Wnt-1 mouse model. In these bigenic mice, both iFGFR1 and iFGFR2 activation resulted in a dramatic enhancement of mammary tumorigenesis. more...
