NO2 study

(Submitter supplied) NO effects on gene expression, independent of cGMP, were examined globally. Differentiated human U937 cells that lack soluble guanylate cyclase were exposed to Snitrosoglutathione, a NO donor, or glutathione without or with dibutyryl-cAMP (Bt2cAMP). NO regulated 110 transcripts that annotated disproportionately to the cell cycle and cell proliferation (47/110, 43%) and more frequently than expected contained AU-rich, post-transcriptional regulatory elements (ARE). more...

Nitric oxide effect on soluble guanylate cyclase deficient cells

Analysis of soluble guanylate cyclase-deficient U937 cells exposed to the nitric oxide (NO) donor S-nitrosoglutathione. Cells treated in the presence of dibutyryl-cAMP (Bt2cAMP) to block signaling due to NO-induced decreases in cAMP. Results provide insight into the cGMP-independent effects of NO.

SAGE analysis of cardiac fibroblasts isolated from adult mouse heart

(Submitter supplied) Cardiac fibroblasts were isolated from 3-month old C57BL/6 mice. RNA was prepared from quiescent, serum-starved cardiac fibroblasts in passage 15. A total number of 118654 tags was sequenced. After removal of duplicate ditags and linker-derived sequences 110169 tags remained.

Live cell monitoring of periodic gene expression in synchronous human cells identifies forkhead genes involved in cell cycle control

(Submitter supplied) We report the genome wide DNA binding patterns of wild type FOXK1 in asynchronous HeLa cells using chromatin precipitation followed by high-throughput sequencing (ChIP-seq). We find that FOXK1 is bound to the promoter of a number of cell cycle genes including the E2F binding partner TFDP1. This study is the first reported study of FOXK1 binding on a genome-wide scale in HeLa cells.

Expression data generated by full-length and truncated syndecan-1 overexpression in B6FS fibrosarcoma cell line

(Submitter supplied) To dissect the functions of syndecan-1 in the nucleus, and separate them from functions related to the cell-surface, we transfected fibrosarcoma cells with two constructs: one encoding the full-length syndecan-1, which translocates to the nucleus and another encoding syndecan-1 lacking the RMKKK nuclear localization signal with hampered nuclear translocation. Using this model system, we combined global transcriptomic and proteomic approaches in order to disclose the molecular mechanisms governing the nuclear translocation and its related functions. more...
