Azithromycin-treated PAO1 vs untreated PAO1

(Submitter supplied) Experimental Design Type of experiment: In order to evaluate the influence of subinhibitory concentrations of the macrolide antibiotic Azithromycin (AZM) on the global Pseudomonas aeruginosa gene expression, we performed a comparative gene expression analysis of AZM-treated versus untreated P. aeruginosa PAO1 cultures. Experimental factors: P. aeruginosa PAO1 cultures with and without addition of 2 µg/ ml AZM were grown until early stationary phase. more...

Azithromycin effect on Pseudomonas aeruginosa

Analysis of Pseudomonas aeruginosa cultures exposed to the macrolide azithromycin (AZM). P. aeruginosa infection is the major cause of morbidity and mortality in cystic fibrosis patients. Results provide insight into mechanisms underlying the beneficial role of AZM therapy in infection control.

High-throughput 3-prime end RNA-seq of Pseudomonas aeruginosa response to Azithromycin

(Submitter supplied) P. aeruginosa produces serious chronic infections in hospitalized patients and immunocompromised individuals, including cystic fibrosis patients. The molecular mechanisms by which P. aeruginosa responds to antibiotics and other stresses to promote persistent infections may provide new avenues for therapeutic intervention. Azithromycin (AZM), an antibiotic frequently utilized in cystic fibrosis treatment, is thought to improve clinical outcomes through a number of mechanisms including impaired biofilm growth and quorum sensing in P. more...

Investigations of the effects of subMIC antibiotics on the P. aeruginosa transcriptome

(Submitter supplied) Antibiotics in low concentrations may have an array of effects. We have exposed growing P. aeruginosa cells to different classes of antibiotics in order to investigate this hypothesis. We have used DNA microarrays to investigate the effect of low concentrations of antibiotics (azithromycin, ceftazidime and ciprofloxacin) on the gene expression of P. aeruginosa aiming to elucidate general trends. Using RNA Seq to define the regulon of the Pseudomonas aeruginosa transcription factor Anr in low-oxygen colony biofilms.

(Submitter supplied) Purpose : The goal of this study was to use RNA Seq to define the regulon of the transciption factor Anr by comparing global transcriptional profiles of Pseudomonas aeruginosa strain PAO1 and a clinical isolate with their isogenic ∆anr mutants, grown in colony biofilms at 1% oxygen. Methods : mRNA profiles were generated for laboratory strain PAO1 and for a clinical isolate J215, as well as for ∆anr derivatives of each strain, in duplicate, by deep sequencing. more...

Expression data from Pseudomonas aeruginosa exposed to a low concentration of Azithromycin

(Submitter supplied) Low-dose macrolides are effective therapy in patients with chronic lung infections, but the mechanisms of action are unclear. We compared global gene expression profiles between P.aeruginosa with and without low-dose Azithromycin (AZM) to study why the low-dose macrolide therapy is effective for cystic fibrosis and diffuse panbronchiolitis. Azithromycin effect on Pseudomonas aeruginosa PAO1

Analysis of PAO1 cultures exposed to up to 2.0 ug/ml of the macrolide azithromycin (AZM) for 9 hours. Low-dose macrolides are effective therapy in patients with chronic lung infections such as those caused by P. aeruginosa. Results provide insight into mechanism of action of AZM against infection.

Differential gene expression from azithromycin and SMM treated human airway epithelium

(Submitter supplied) Microarrays were used to evaluate the effects of azithromycin and an inflammatory stimulus (SMM) on human airway epithelium. Effects of azithromycin treatment were evaluated at 6, 24 and 48 hours. Effects of SMM were evaluated at 6 and 24 hours. In addition, pretreatment with azithromycin was used to evaluate the modulatory effects on SMM-induced inflammation. SMM=supernatant from microcorpulent material from human cystic fibrosis airways. more...

Steady-state hydrogen peroxide induces glycolysis via metabolic reroute in P. aeruginosa and S. aureus

(Submitter supplied) GAPDHs from human pathogens S. aureus and P. aeruginosa can be readily inhibited by ROS-mediated direct oxidation of their catalytic active cysteines. Because of the rapid degradation of H2O2 by bacterial catalase, only steady-state but not one-dose treatment of H2O2 induces rapid metabolic reroute from glycolysis to pentose phosphate pathway (PPP). We conducted RNA-seq analyses to globally profile the bacterial transcriptomes in response to a steady level of H2O2, which reveals profound transcriptional changes including the induced expression of glycolytic genes in both bacteria. more...
