Folic Acid study in HT-29 cells

(Submitter supplied) Low intracellular folate levels diminish the growth rate of HT-29 human colon cancer cells. This is accompanied by a metabolic shift from cytosolic glycolysis towards mitochondrial oxidative phosphorylation, as demonstrated by a lower lactate production and an increased mitochondrial oxygen consumption rate. Folic acid deficiency effect on colon cancer cells

Analysis of HT-29 colon cancer cells cultured in pteroylglutamic acid or methyl-tetrahydrofolate at concentrations of 10 or 100 ng/ml each. Studies suggest that folate deficiency can have an inhibitory effect on the progression of established colonic tumor cells.

RIKILT Human Oligo Array

(Submitter supplied) The 10K MWG human oligoset together with additional 50-mer oligo probes involved in energy metabolism were in-house printed on GAP II coated slides (Corning), using the Microgrid II arrayer (BioRobotics Ltd, Cambridge, UK). After printing, microarrays were allowed to dry at room temperature. The oligo arrays were rehydrated and immobilized by UV cross-linking (120 MJ of UV energy). The microarray slides were then prewashed in the following order to reduce background; 20 minutes at 55C with 2x SSC/0.2% SDS, 5 minutes at room temperature (RT) with 0.2x SSC and 3 minutes at RT with demineralized water. more...
