Regulatory Mechanisms That Prevent Re-initiation of DNA Replication Can Be Locally Modulated at Origins by Nearby Sequence Elements

(Submitter supplied) Eukaryotic cells must inhibit re-initiation of DNA replication at each of the thousands of origins in their genome because re-initiation can generate genomic alterations with extraordinary frequency. To minimize the probability of re-initiation from so many origins, cells use a battery of regulatory mechanisms that reduce the activity of replication initiation proteins. Li UCSF yeast 14062 v1.0

(Submitter supplied) PCR products representing ORF and intergenic yeast genomic sequences were amplified. Products were spotted on poly-L-lysine coated glass slides using a linear servo arrayer from the DeRisi Lab at UCSF. Blast analysis of oligos sequences was used to determine the position of the final PCR product.
