Mechanisms of Pancreatic Acinar Cell Neoplasia Associated with Gene Expression Changes caused by Dietary Exposure of Rats to Ammonium perfluorooctanoate, Wyeth 14,643 or Diethylhexyl phthalate

(Submitter supplied) To identify putative procarcinogenic gene changes associated with pancreatic acinar cell neoplasia in rats exposed to peroxisome proliferator activated receptor alpha agonists, we performed transcription profiling in the pancreas of rats fed diets containing the pancreatic carcinogen Wyeth 14,643 (Wy) at procarcinogenic (50ppm) and non-carcinogenic (20ppm) doses for 1, 7, 28 and 90 days. The effects of these treatments were compared to those of ammonium perfluorooctanoate (APFO) 300 ppm, an agent shown to induce pancreatic acinar cell adenomas in rats, and diethylhexylphthalate (DEHP) 12,000 ppm, an agent not shown to produce pancreatic acinar cell tumors. more...

Gene expression of rat uterine endometrial stromal cells treated with ethanol (vehicle) or 0.1 nM progestin R5020

(Submitter supplied) Although non-genomic steroid receptor pathways have been studied over the past decade, little is known about the direct gene expression changes that take place as a consequence of their activation. Progesterone controls proliferation of rat endometrial stromal cells during the peri-implantation phase of pregnancy. We showed that picomolar concentration of progestin R5020 mimics this control in UIII endometrial stromal cells via ERK1-2 and AKT activation mediated by interaction of Progesterone Receptor (PR) with Estrogen Receptor beta (ERb) and without transcriptional activity of endogenous PR and ER. more...

Cultured OEC versus cultured SC and versus native OEC

(Submitter supplied) This microarray study aimed at identifying the differences in the global gene expression growth program of adult cultured olfactory ensheathing cells (cOEC) (from the olfactory bulb) versus adult cultured Schwann cells (SC) (from the sciatic nerve) and versus adult OEC directly dissected from the olfactory nerve layer (nOEC). The aim of the comparison between cOEC and SC is to define intrinsic molecular differences that distinguish cOEC from SC (both cell types support neuronal regeneration). more...

Aortic gene expression profile of rats exposed in utero to maternal diabetes

(Submitter supplied) Epidemiologic studies have clearly identified modifications of fetal environment as a risk factor for the development of cardiovascular diseases and of metabolic syndrome in adulthood. In our experimental model, rats exposed to maternal diabetes develop a volume-dependent hypertension as early as 6 month of age. In order to determine if the development of hypertension results from an abnormal vascular fetal programming, gene expression profile of thoracic aorta were studied in adult rats exposed in utero to maternal hyperglycemia.

Identification of flap structure specific endonuclease 1 as a factor involved in LTM formation of aversive learning

(Submitter supplied) We proposed that DNA recombination/repair processes play a role in memory formation. Here, we used microarray analysis of rat amygdala genes to identify possible DNA recombination/repair factors involved in memory consolidation of conditioned taste aversion (CTA). Among the genes that showed statistically significant differential expression, we identified fen-1, encoding a flap-structure specific DNA endonuclease. more...

Expression Profiling of Genes Concerning to Protective Effect in Heart by Benidipine Hydrochloride.

(Submitter supplied) Benidipine hydrochloride is a long-acting calcium channel blocker and is used for the treatment of hypertension and angina pectoris. The cardioprotective effects of benidipine have been shown in several animal and clinical studies. We examined the effect of the dihydropyridine calcium channel blocker (CCB) benidipine on the gene expression profile in heart by DNA microarray. Dahl salt-sensitive (DS) rats were fed with normal diet (0.19% NaCl) or a high-salt diet (8% NaCl) from 7 weeks of age. more...

Acetaminophen-induced gene expression profiles in sandwich-cultured primary rat hepatoctyes

(Submitter supplied) The frequent use of rodent hepatic in vitro systems in pharmacological and toxicological investigations challenges extrapolation of in vitro results to the situation in vivo and interspecies extrapolation from rodents to humans. The toxicogenomics approach may aid in evaluating relevance of these model systems for human risk assessment by direct comparison of toxicant-induced gene expression profiles and infers mechanisms between several systems. more...

The acute effects of methoxyacetic acid exposure on testis gene expression

(Submitter supplied) We evaluated the possible mechanisms by which 2-methoxyacetic acid (MAA) disrupts spermatogenesis using high density microarrays. Levels of mRNA transcripts were determined in total RNA isolated from testes of MAA-treated (650 mg/kg i.p.) or concurrent control rats sacrificed 4, 8, 12 or 24 hrs post exposure (PE). Gene Expression Analysis of the Rat Retina during Maturation

(Submitter supplied) Recently we reported that the rat inner retina undergoes significant functional changes during maturation. Aiming to gain knowledge on additional aspects of retinal development and maturation, we used the microarray system to monitor gene expression patterns in the rat retina at ages 5, 11, and 20 weeks. The analysis revealed the expression of many well-documented retinal genes as well as a high number of non–annotated genes. more...

Global gene expression analysis of quercetin bioactivity in cultured rat cardiomyocytes

(Submitter supplied) Despite extensive studies, the fundamental mechanisms responsible for the development and progression of heart failure have not yet been fully elucidated. Recent experimental and clinical studies have suggested that reactive oxygen species (ROS) play a major pathological role. Quercetin, a major flavonoid present in the human diet, has been studied widely and its biological properties are consistent with its protective role in the cardiovascular system. more...

Global gene expression profiling on renal scarring in rat model of pyelonephritis

(Submitter supplied) Renal scarring is a serious complication of chronic pyelonephritis due to vesicoureteral reflux, and requires specific biomarker for indication of early intervention. We investigated global expression profile of kidney during renal scarring formation using rat pyelonephritis model. An inoculum of the DS-17 strain Escherchia coli was injected directly into renal cortex of Wister rat. Histologically, renal scarring developed during 3 and 4 weeks after injection. more...

Modulation of cardiomyocyte gene expression by n-3 PUFA supplementation

(Submitter supplied) Despite the recognized protective effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in cardiovascular diseases, and the demonstration of the control of gene expression by polyunsaturated fatty acids (PUFAs), the effects of these n-3 fatty acids on the whole genoma has never been investigated in cardiac cells. Using rat arrays, the effects of Eicosapentaenoic acid (EPA) and Docosahexaenoic acid (DHA) supplementation on the global gene expression profile were evaluated in cultured neonatal rat cardiomyocytes. more...
