A Marfan syndrome gene expression phenotype in cultured skin fibroblasts

(Submitter supplied) Background: Marfan syndrome (MFS) is a heritable connective tissue disorder caused by mutations in the fibrillin-1 gene. This syndrome constitutes a significant identifiable subtype of aortic aneurysmal disease, accounting for over 5% of ascending and thoracic aortic aneurysms. Results: We used spotted membrane DNA macroarrays to identify genes whose altered expression levels may contribute to the phenotype of the disease. more...

Translation State Array Analysis of Serum Stimulated Human Endothelial Cells

(Submitter supplied) Confluent human umbilical vein endothelial cells (HUVECs) were exposed to histamine (100 mM) for 2 hours. Ribosomal profiling via gradient centrifugation and fractionation was used to separate monosome, or under-translated, and polysome, or actively translated, mRNA species that were then used to probe cDNA arrays, a process known as Translation State Array Analysis (TSAA). Four samples were obtained from these experiments, control monosome, control polysome, histamine monosome and histamine polysome. more...

Trascriptional profiling of angiogenic inhibitors

(Submitter supplied) To examine angiogenesis inhibitor-regulated pathways, the alteration in gene expression of human microvascular endothelial cells after a 4-hour treatment with seven chemically different inhibitors has been analyzed. Specifically, this series describes the expression profiles following treatment with either: 1. recombinant interferon-alpha (IFN-alpha); 2. pigment epithelium derived factor (PEDF); 3. all trans-retinoic acid (RA); 4. thrombospondin 1 (TSP-1); 5. TNP-470 (TNP-470); 6. endostatin (ENDO); 7. angiostatin (ANGIO). To ensure that cDNA products were proportional to the initial gene expression pattern, 33P-labelled probes from oligo d(T) primed reverse transcription of 5 microg RNA were generated for each. This, followed by second strand synthesis, was used to hybridize the nylon filters (GF211 ) comprising a total of 4132 genes and EST’s. Autoimmune family study

(Submitter supplied) Patients with autoimmune disorders exhibit highly reproducible gene expression profiles in their peripheral blood mononuclear cells. These signatures may result from chronic inflammation, other disease manifestations, or may reflect family resemblance. To test the latter hypothesis, we determined gene expression profiles in unaffected first-degree relatives of individuals with autoimmune disease. Gene expression profiles in unaffected first-degree relatives resembled the profiles found in individuals with autoimmune diseases. more...

Gene Expression Signature Shared in Autoimmune Diseases Not in Unaffected Family Members

(Submitter supplied) Even though autoimmune diseases are heterogeneous, believed to result from the interaction between genetic and environmental components, patients with these disorders exhibit reproducible patterns of gene expression in their peripheral blood mononuclear cells. A portion of this gene expression profile reflects family resemblance rather than the actual presence of an autoimmune disease. Here we wanted to identify that portion of this gene expression pattern that is independent of family resemblance and determine if it is a product of disease duration, disease onset, or other factors. more...

Allergy response

(Submitter supplied) Background Atopic diseases, resulting from hypersensitivity to a wide variety of allergens, affect 10-20% of the population. Immunotherapy is an effective treatment for atopic diseases, but its mechanisms are not fully understood. Objective We studied gene expression profiles in the peripheral blood mononuclear cells and examined whether the individuals with allergic rhinitis have a unique gene expression profile and how the immunotherapy affect the gene expression profiles. more...
