Peritoneal fluid from subjects with endometriosis regulates gene expression in cultured endometrial cells

(Submitter supplied) Purpose: This study was undertaken to evaluate the effect of peritoneal fluid from subjects with differing stages of endometriosis on gene expression in endometrial stromal cells. Methods: Peritoneal fluid from subjects with minimal, moderate, and severe stages of endometriosis or without endometriosis (controls) was collected, filtered and separated from peritoneal cells. Telomerase – immortalized human endometrial stromal cells (T-HESC) were treated with the peritoneal fluid samples for 48 hours. more...

Sex-related differences in lactotroph tumor aggressiveness are associated with a specific gene-expression signature and genome instability

(Submitter supplied) we aimed to explore the molecular importance of the estrogen pathway through a detailed analysis of the transcriptomic profile of lactotroph tumors from 20 men and 10 women. Transcriptional changes of influenza A/Hong Kong/1/68-H3N2 (HK68) infected A549 lung epithelial cells

(Submitter supplied) Typically viral infections make the host more susceptible to subsequent bacterial infections by mechanisms that are incompletely defined. To identify host changes that occur in response to HK68 infection, gene expression of A549 lung epithelial cells were identified. Changes included an increase in transcripts encoding proteins with fibronectin-type III (FnIII) domains, such as fibronectin (Fn), tenascin N (TNN), and tenascin C (TNC). more...

Gene expression profiling of patients affected by first acute myocardial infarction (FAMI)

(Submitter supplied) Background: Myocardial infarction (MI) is the single most critical event in coronary disease. Platelets partecipate in processes that precipitate acute MI, lack nuclear DNA but retain megakaryocyte mRNAs, thus their transcriptome could provide information preceding the coronary event. Our aim was to obtain a gene expression atlas of platelets from patients with their very first acute MI (FAMI) in order to identify potential markers of precipitating factors. more...

The gene expression analysis of human astrocyte exposure to anti-suprabasin antibody: identification of a novel antibody for neuropsychiatric systemic lupus erythematosus

(Submitter supplied) [Objectives]: Neuropsychiatric systemic lupus erythematosus (NPSLE) is often difficult to diagnose and distinguish from those of other diseases, because no specific antibodies have yet been detected. [Methods]: We developed a novel proteomic strategy for identifying and profiling antigens in immune complexes (ICs) in the cerebrospinal fluid (CSF) of 26 NPSLE patients. We performed in vitro experiments using astrocytes and analyzed functional change by microarray. more...

MIA gene family-related signaling in oral squamous cell carcinoma cells

(Submitter supplied) Melanoma inhibitory activity (MIA) gene family is novel tumor-associated molecules. Although MIA gene family has several tumor progressive and/or suppressive functions, the detailed relevant signaling partners are unclear and investigated. In this study, we investigated the detailed MIA gene family-associated signaling using human oral squamous cell carcinoma cells.

Gene expression profile associated with a cytostatic does of mifepristone in ovarian cancer cells by microarray analysis

(Submitter supplied) Goal of the experiment: To examine differential gene expression in three ovarian cancer cell lines in the absence and presence of mifepristone. Brief description of the experiment: Hypothesis: The gene expression profile associated with a cytostatic response to Mifepristone will be similar between ovarian cancer cells regardless of their p53 status or sensitivity to Cisplatin. Aim: Evaluate the modifications in gene expression profile in ovarian cancer cells in response to a cytostatic concentration of Mifepristone against those in logarithmic phase of growth (Vehicle). more...

Analysis of gene expression changes when KLF6 is overexpressed in HepG2 cells

(Submitter supplied) KLF6 was overexpressed using an adenoviral delivery system. Overexpression was confirmed at 12, 24, 48 and 72 hours post infection by RT-PCR and Western Blot analysis. Codelink Whole genome microarrays were used to analyse changes in expression resulting from KLF6 overexpression.

A PU.1 suppressive target gene, metallothionein-1G, inhibits retinoic acid-induced NB4 cell differentiation

(Submitter supplied) We recently revealed that myeloid master regulator PU.1 directly represses metallothionein (MT)-1G through its epigenetic activity, but the functions of MT-1G in myeloid differentiation remain unknown. To clarify this, we established MT-1G-overexpressing acute promyelocytic leukemia NB4 (NB4MTOE) cells, and investigated whether MT-1G functionally contributes to all-trans retinoic acid (ATRA)-induced NB4 cell differentiation. more...

Role of calcium/calmodulin-dependent kinase type IV in podocyte function in lupus nephritis

(Submitter supplied) Kidney podocytes and their slit diaphragms contribute to prevent urinary protein loss. T cell from patients with systemic lupus erythematosus display increased expression of calcium/calmodulin kinase IV (CaMKIV). Here we evaluated the functional role of CaMKIV in lupus nephritis (LN) using kidney biopsy specimens and human podocyte cell line (AB8/13). We found that exposure of podocytes to IgG from LN patients resulted in entry of IgG into the cytoplasm. more...

TNFalpha and IL1beta stimulate differential gene expression in endometrial stromal cells

(Submitter supplied) Cytokines are implicated in the development of inflammatory diseases such as endometriosis. This project was designed to test the hypothesis that specific cytokines that are secreted by macrophages, such as tumor necrosis factor α (TNFα) and interleukin 1 beta (IL1β), cause gene expression changes in endometrial stromal cells. Telomerase-immortalized human endometrial stromal cells (T-HESC) were treated with TNFα (5ng/ml) ± IL1β (1ng/ml). more...

Effects of equol on gene expression in female cynomolgus monkey iliac arteries

(Submitter supplied) Objective: To examine the effects of the phytoestrogen metabolite, equol, on gene expression in the monkey iliac artery. Design: Eight ovariectomized cynomolgus monkeys were on a high fat diet for 6.5 years. The left iliac artery was biopsied prior to randomization to equol (0.0237 g/100 g chow, n=4) or vehicle (n=4) for 8 months. The right iliac artery was obtained at necropsy. Gene expression in the iliac arteries in response to equol was determined by DNA microarray and confirmed by real time RT-PCR. more...

Microarray profiling of mononuclear peripheral blood cells to identify novel candidate genes related to chemoradiation response in rectal cancer

(Submitter supplied) Peripheral blood mononuclear cells have emerged recently as pathology markers of cancer and other diseases, making possible their use as therapy predictors. Furthermore, the importance of the immune response in radiosensivity of solid organs led us to hypothesized that microarray gene expression profiling of peripheral blood mononuclear cells could identify patients with response to chemoradiation in rectal cancer. more...
