Effects of AEBP1 knockdown on gene expression profiles in human skeletal muscle myoblasts

(Submitter supplied) We recently identified adipocyte enhancer binding protein 1 (AEBP1) as a novel cancer stroma-associated gene and reported that AEBP1 activates cancer-associated fibroblasts in oral cancer. In this study, we show that AEBP1 is negatively associated with skeletal muscle cell differentiation. We found that AEBP1 is downregulated during the diffraction of human skeletal muscle myoblasts (HSMMs). Microarray analysis revealed that knockdown of AEBP1 significantly upregulated a number of genes associated with myogenesis in HSMMs. more...

Inflammation promotes stomach epithelial defence by stimulating the secretion of antimicrobial peptides in the mucus

(Submitter supplied) Mucus is a protein-based gel secreted by specialized epithelial cells that protects the gastrointestinal mucosa from microorganism attack and irritating agents. Experiments in mice have demonstrated that when the mucosa is infected or inflamed, the mucus becomes enriched with broad-spectrum bactericidal compounds known as antimicrobial peptides (AMPs). Although AMP gene expression has been identified in both immune and epithelial cells, how inflammation regulates AMP secretion in the mucus remains unclear, and the efficacy of AMP-enriched mucus in defending against microorganisms has not been assessed. more...

Analysis of gene expression changes in lipid droplet formation of human liver cancer cell HepG2

(Submitter supplied) Fatty liver is a condition of excessive triglyceride accumulation in hepatocytes. Additionally, hepatocytes showed a high degree of fat droplet accumulation during excessive alcohol consumption and metabolic syndrome. However, the molecular mechanisms involved in fat droplet formation remain unknown. This study used an in vitro fatty liver formation model of the human liver cancer cell line HepG2 to comprehensively search for fat droplet formation-related genes whose expression changes during fat droplet formation. more...

Dual targeted extracellular vesicles regulating oncogenic genes in pancreatic cancer [Human Array]

(Submitter supplied) Pancreatic ductal adenocarcinoma (PDAC) tumors carry multiple gene mutations and respond poorly to treatments. There is currently an unmet need for drug carriers that can deliver multiple gene cargoes to high solid tumor burden like PDAC. Here, we report a dual-targeted extracellular vesicle (EV) carrying high loads of RNA that effectively suppresses large PDAC tumors in mice. The EV surface contains a CD64 protein that has a tissue targeting peptide and a humanized monoclonal antibody. more...

Transcriptomic profiling reveals differential cellular response of copper oxide nanoparticles and polystyrene nanoplastics in perfused human placenta

(Submitter supplied) Concerns about the potential risks to human health due nanoparticulate pollution have been emerging. However, the risks to sensitive populations, such as pregnant individuals and their unborn children are poorly characterised. With increasing evidence of environmental particles passing the placenta, their potential adverse effects of on pregnancy and fetal development need to be assessed. Here, we investigated the impact of copper oxide (CuO) and polystyrene (PS) nanoparticle exposure on gene expression in ex vivo perfused human placental tissue.

PRC1.6 molecule L3MBTL2 protects MYCN-amplified neuroblastoma cells via epigenetic gene suppression

(Submitter supplied) Polycomb-group complexes are evolutionarily conserved epigenetic machineries that regulate stem cell fate decisions/development and are also implicated in tumorigenesis mainly by histone modification. PRC1 complexes mediates ubiquitylation of histone H2A on lysine 119 and consists of PRC1.1 to PRC1.6 complexes. Here, we studied the functional roles of a PRC1.6 molecule L3MBTL2 in neuroblastoma (NB) cells. more...

Effects of AEBP1 knockdown on gene expression profiles in cancer-associated fibroblasts

(Submitter supplied) We identified adipocyte enhancer binding protein 1 (AEBP1) as a novel cancer-associated fibroblast (CAF) marker in oral squamous cell carcinoma (OSCC). To study the function of AEBP1 in CAFs, we knocked down AEBP1 in CAFs isolated from surgically resected primary OSCCs. We found that genes associated with cell cycle were significantly affected by AEBP1 knockdown in CAFs.

Transcriptome analysis of human maturing macula

(Submitter supplied) The macula is an essential region of the retina responsible for visual acuity. Abnormalities in development of the fovea at the center of the macula lead to severe diseases. To identify macula-specific differentially expressed genes in maturing stage of macular development, we compared the gene expression profiles of the macular and peripheral region of the pediatric retina.

Impact of sequential exposures to allergens and ultrafine particles on human bronchial epithelial BEAS-2B cells at the air liquid interface

(Submitter supplied) BEAS-2B cells, at air liquid interface, were exposed to birch pollen extract or house dust mite extract in a cloud chamber and, later, to UFP rich combustion aerosols in an in vitro exposure system. As control the same exposure was performed without allergen containing extracts. The goal was to understand the effect of allergenic pre-exposure to a UFP rich combustion aerosol exposed cells and their effect on allergic sensitization, using an established model that mimics more closely real life exposures.

PDGF receptors and signaling are required for 3D-structure formation and differentiation of human iPSCs-derived hepatic spheroids

(Submitter supplied) Human iPSC-derived liver organoids (LO) or hepatic spheroids (HS) have attracted widespread interest and the numerous studies on them have recently provided various production protocols. However, the mechanism by which the 3D structures of LO and HS are formed from the 2D-cultured cells and the mechanism of the LO and HS maturation remain largely unknown. In this study, we demonstrate that PDGFRA is specifically induced in the cells that are suitable for HS formation and that PDGF receptors and signaling are required for HS formation and maturation. more...

Identification and characterization of pluripotency associated lncRNAs in human iPS cells

(Submitter supplied) Induced pluripotent stem cells (iPSCs) are an attractive source of stem cells for many applications like patient-specific cell therapy, disease modeling or drug screening. Generation of iPSCs by cellular reprogramming is a complex and highly inefficient process that involves a high number of genetic and epigenetic modifications most of them unknown. Those genetic modifications include both coding and non-coding genes like long non-coding RNAs (lncRNAs). more...

PRC2 molecule EED is a target of epigenetic therapy of neuroblastoma

(Submitter supplied) Epigenetic modification by polycomb repressive complex (PRC) molecules appears to have a role in tumorigenesis and aggressiveness of neuroblastoma (NB). Embryonic Ectoderm Development (EED) is a member of PRC2 complex and binds the H3K27me3 mark deposited by EZH2, via propagation on adjacent nucleosomes. Here we studied the molecular roles of EED in MYCN-amplified neuroblastoma cells by using EED-knocked down shRNAs, EED-knocked out sgRNAs, and EED small molecule inhibitor EED226. more...
