Gene expression in brain following stroke

(Submitter supplied) We have used microarrays to investigate the changes in gene expression at various times after stroke. Our findings reported that gene expression screening can detect known and unknown transcriptional features of stroke. Keywords: time course, disease state analysis

Myocardial gene expression of hibernating and control tissue from patients with ischemic left ventricular dysfunction

(Submitter supplied) Objectives: While cardiac scar tissue is damaged irreversibly, hibernating myocardium is characterized by reversible contractile dysfunction. Limited data are available in humans regarding the molecular biology of hibernating myocardium. The aim of this study was to identify new molecular mechanisms distinctive for human hibernating myocardium by gene expression analysis. Results: Of 4,171 transcripts examined, we identified 86 to be differentially expressed. more...

Expression of inflammatory host genes in Chlamydia trachomatis infected human monocytes

(Submitter supplied) Messenger RNA of C trachomatis infected monocytes of the three healthy donors was compared with the corresponding mock-infected samples. For mock infection, SPG buffer was used instead of the C trachomatis suspension, all other procedures were performed identically. Cells were resuspended in solution D and total RNA was extracted by application of phenol:chloroform 5:1 (pH 4.5, Ambion, Austin, TX) followed by precipitation in isopropanol at -80°C for 1 hr and incubation with RQ1 DNase (Promega, Madison, WI) at 37 °C for 1 hr. more...

Acute and Chronic Environmental Tobacco Smoke Condensate Exposure to Primary Human Bronchial Epithelial Cells

(Submitter supplied) The goal was to assess global gene expression changes in primary human bronchial epithelial cells exposed to environmental tobacco smoke (ETS) condensate. ETS-C was standardized by HPLC analysis and two timepoints of exposure in two different donor bronchial epithelial cell populations were assessed. These findings demonstrate that even short exposure (4.5 h) to ETS is sufficient to induce a stress response, as reflected by decreased antioxidant levels, induced HSP family members, and modulation of the family of glutathione metabolism enzymes in primary human lung cells. more...

Genes modulated by DNA demethylation in UM1 cells

(Submitter supplied) Ornithine decarboxylase antizyme 1 mediates DNA demethylation. Using a human oral cancer cell line, UM1, genes induiced by DNA demethylation were screened. To verify the methylation status of those induced genes, the gene expression profile was compared with that of 5-Aza-2’-deoxycytidine treated sample. Keywords: DNA demthylation, ornithine decarboxylase antizyme 1, 5-Aza-2’-deoxycytidine

HEVEC dedifferentiation

(Submitter supplied) A double screen has been carried out by comparing freshly isolated HEVEC (D0-Hevec) from human tonsils to freshly purified HUVEC (D0-Huvec), and to dedifferentiated HEVEC (D2-Hevec) grown ex vivo for two days, using three Clontech commercial nucleotide arrays (Atlas Human Cardiovascular, 1.2 I and 1.2II Arrays) Keywords = freshly isolated HEVEC expression profile Keywords = HEVEC dedifferentiation ex vivo Keywords: other
