Description

In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Loss-of-function variants in SDHB are known to be pathogenic (PMID: 19454582, 19802898). This variant has not been reported in the literature in individuals with SDHB-related conditions. This variant is a complex sequence change that deletes genomic region encompassing part of exon 3 and inserts another 14 nucleotides at the intron 3-exon 4 boundary (c.208_287-1359delins14) of the SDHB gene. It is expected to create a premature translational stop signal and result in an absent or disrupted protein product.
